荔枝核总黄酮对大鼠肝纤维化PDGF、TNF-α因子的影响

【摘要】:目的:探讨荔枝核总黄酮(total flavone from Litchi Chinensis Sonn,TFL)对大鼠肝纤维化PDGF及TNF-α因子的影响。方法:SD雄性大鼠60只,,随机分为空白组、模型组、水飞蓟宾(SIL)组(50mg kg-1d-1)、TFL高剂量组(400mg kg-1d-1)、TFL中剂量组(200mg kg-1d-1)、TFL低剂量组(100mg kg-1d-1)六组。采用二甲基亚硝胺DMN腹腔注射制备SD大鼠进行肝纤维化造模,造模4周,造模同时SIL、TFL等四组灌胃给药干预,6周后处死大鼠,观察大鼠肝纤维化水平,用放射免疫法测定模型组、SIL组和TEL组大鼠血清中层粘蛋白(LN)、透明质酸(HA)及Ⅲ型前胶原(PCⅢ)水平,用HE染色法和Masson染色观察大鼠肝组织镜下病理变化纤维化水平,采用PCR法检测SIL组和TFL组PDGF、TNF因子表达水平变化,用SPSS11.0软件对数据进行统计分析。结果:与模型组大鼠比较, SIL和TFL大、中剂量治疗后大鼠血清中HA、LN和PCⅢ的水平显著降低(P<0.01);肝组织内纤维化程度降低,肝小叶结构趋于正常;TFL大、中剂量给药组和水飞蓟素组血清PDGF及TNF-α水平均显著低于模型组(P<0.05);两组大鼠肝组织PDGF及TNF-α表达明显降低(P<0.05)。TFL中剂量给药组与对照组比较血清HA、LN和PCⅢ的水平、肝纤维化程度、PDGF及TNF-α表达水平均无明显差异(P>0.05)。结论:TFL可以下调二甲基亚硝胺肝纤维化大鼠的PDGF、TNF-α细胞因子表达水平,具有明显的抗大鼠肝纤维化作用。
[Abstract]:Objective: to investigate the effects of total flavonoids (total flavone from Litchi Chinensis Sonn,TFL on PDGF and TNF- 伪 in rat liver fibrosis. Methods: sixty male SD rats were randomly divided into blank group, model group and silybin (SIL) group (50mg kg-1d-1), TFL high dose group (400mg kg-1d-1), TFL medium dose group) 200mg kg-1d-1), TFL low dose group (100mg kg-1d-1) group. SD rats were induced by intraperitoneal injection of dimethylnitrosamine (DMN) to model liver fibrosis for 4 weeks. Four groups of rats, such as SIL,TFL, were given intragastric administration of dimethylnitrosamine. After 6 weeks, the rats were killed to observe the level of hepatic fibrosis.
