搭载klotho蛋白热敏型聚合凝胶在内镜下治疗胃溃疡

【摘要】:目的:1、探索klotho蛋白是否对GECs(胃上皮细胞)增殖率和迁移率有促进作用;2、制作一款包裹klotho蛋白能用于内镜下的热敏型聚合凝胶并对材料进行评估;3、通过动物实验验证聚合凝胶对胃溃疡的愈合是否有促进作用。方法:1、细胞增殖实验(EDU)和细胞迁移实验(Transell):将对数期生长的GECs分为klotho蛋白组和对照组,klotho蛋白组添加klotho,观察两组细胞增殖率和迁移率有无差异。2、聚合凝胶的制作及评估:以β-环糊精包裹klotho蛋白的制成纳米微球,用壳聚糖、F-127、甘油等材料制作成一种温度敏感性凝胶,纳米微球与凝胶混合均匀后通过扫描电镜观察,并测定生物材料中klotho蛋白的缓释速度。3、胃潴留实验:将8只C57小鼠分为2组,对照组和实验组,分别灌入含1%亚甲蓝的生理盐水、klotho蛋白凝胶,在0、30、60、120分钟后取胃,通过活体荧光成像系统检测荧光强度。4、动物实验:将60只SD大鼠随机分为生理盐水组,空白凝胶组,奥美拉唑组、klotho蛋白凝胶组4组。各组大鼠成模后每天分别用生理盐水、空白凝胶、奥美拉唑、Klotho蛋白凝胶灌胃1次治疗。在治疗的第1、3、7天每组随机处死5只大鼠取材,测量溃疡面积及新生毛细血管数量。溃疡标本切片后行HE染色、PAS染色、PNCA染色,观察各组组织学变化情况。结果:1、细胞实验:klotho蛋白组的增值率是对照组的2.3倍,细胞迁移数量是对照组的3.6倍。2、电镜观察结果:纳米微球直径在50-200nm之间,可见纳米微球匀称散布在klotho凝胶上面。3、胃潴留实验:klotho凝胶的荧光的强度在每个时间段都明显高于对照组。4、动物实验:klotho凝胶组愈合情况从肉眼观和组织学角度都要优于各个对照组。结论:1、证明了Klotho蛋白在酸性条件下能够促进GECs的增殖迁移。2、制备的包裹klotho蛋白凝胶能够成功用于内镜下并黏附在胃溃疡部位。3、证实了klotho蛋白凝胶能够成功地通过灌胃器导管作用于大鼠胃溃疡部位,修复溃疡组织。
