烟碱及单侧迷走神经切断对重症急性胰腺炎模型大鼠肺损伤的影响
[Abstract]:Objective to observe the effects of nicotine and vagus nerve transection on (SAP) lung injury in rats with severe acute pancreatitis (SAP). Methods 48 SPF female Wistar rats were randomly divided into 6 groups, 8 rats / group, treated with), SAP saline in (Sham saline group and), SAP nicotine group in (SAP saline group, respectively, and normal control rats were treated with), SAP vagus nerve transection in (Con sham surgery group and), SAP vagus nerve transection (SAP vagotomy group in (SAP sham surgery group. The rat model of SAP was established by retrograde injection of 5% sodium taurocholate into the cholancreatic duct. Nicotine was injected intraperitoneally 30 min before operation or left vagus nerve was cut off 3 days before operation. Rats were killed 12 hours after operation. Pathological changes of pancreas and lung tissue were observed, serum amylase (AMY), lipase (LIPA), enzyme linked immunosorbent assay (Elisa) (ELISA) was used to detect serum IL-6 (IL-6), and lung tissue myeloperoxidase (MPO), immunoblotting (Western blot) was used to detect high mobility group protein B1 (HMGB1) in lung tissue. Results the pathological scores of pancreas and lung tissue, AMY,LIPA,IL-6,MPO and HMGB1 in SAP saline group were higher than those in Sham saline group (P 0.05), and the above indexes in), SAP nicotine group were lower than those in SAP saline group (P 0.05). The above indexes in), SAP sham surgery group were higher than those in Con sham surgery group (P 0.05), and the above indexes in), SAP vagotomy group were higher than those in SAP sham surgery group (P 0.05). Conclusion nicotine can reduce lung injury in SAP by down-regulating IL-6,MPO and HMGB1, and unilateral vagotomy can aggravate lung injury in SAP.
