骨髓间充质干细胞培养上清对肝星状细胞所致肝纤维化治疗的实验研究

  目的肝纤维化是由于各种急慢性炎症刺激肝脏导致损伤修复反应而产生的结果,其中,肝星状细胞(hepatic stellate cells,HSC)的活化被认为是纤维化发生的中心环节,现研究已证实骨髓间充质干细胞(bone marrow mesenchymal stem cells,BMSCs)移植治疗能改善甚至逆转肝纤维化,但其具体机制尚不清楚。本研究通过用人 BMSCs(human bone marrow mesenchymal stem cells,HBMSCs)培养上清诱导HSC,观察诱导后对HSC增殖周期及表达的基质金属蛋白酶-1(matrix metalloproteinase-1,MMP-1)、基质金属蛋白酶-2(matrix metalloproteinase-2,MMP-2)及组织抑制基质金属蛋白酶-1(tissue inhibitors of metalloproteinase-1,TIMP-1)的影响。方法在无菌条件下抽取肝硬化志愿者骨髓5ml,采用密度梯度离心法分离HBMSCs,接种于25cm2塑料培养瓶中培养,收集原代培养7d的HBMSCs培养上清,加入HSC中共培...
