牛磺酸对重症急性胰腺炎大鼠Kupffer细胞P38MAPK信号转导通路的影响

目的用牛磺酸(Taurine)对重症急性胰腺炎(severe acute pancreatitis ,SAP)大鼠进行预处理,采用联合酶原位灌注法分离枯否细胞(Kupffer cells,KCs),检测KCs中P38-丝裂原蛋白激酶(P38 mitogen activated protein kinase, P38MAPK)的蛋白量和其活化水平以及核因子-κB(Nuclear factor-kappaB, NF-κB)的活性,探讨牛磺酸对重症急性胰腺炎大鼠KCs内P38MAPK的影响以及对分泌促炎细胞因子的作用。方法48只雄性SD大鼠采用完全随机法分为:①假手术对照组(sham operation,SO);②重症急性胰腺炎组(SAP);③牛磺酸预处理组(Tau)。SAP模型通过胰胆管逆行注射5%牛磺胆酸钠诱导,造模前Tau组分别于24h和12h从尾静脉内注入牛磺酸(300mmol/L,1.0ml /100g),其它组注入等量的生理盐水。假手术或造模后分别于12h和24h处死动物,检测其血清中AST、ALT和AMS含量;然后采用联合酶原位灌注法分离KCs,采用蛋白免疫印迹法和免疫细胞化学...
