经导管射频消融去肾交感神经术对高血压犬心肌纤维化及miR-133a/CTGF的影响

目的:观察经导管射频消融去肾交感神经术(RDN)对高血压犬心肌纤维化程度、心肌miR-133a、结缔组织生长因子(CTGF)表达的改变,探讨RDN对心肌纤维化的影响及可能的机制。方法:1、用高脂饮食建立高血压犬模型,将28只比格犬(10-12月龄)随机分为对照组(普通饮食,n=6)和模型组(高脂饮食,n=22)。2、建模12周后将建模成功的高血压犬(n=20)随机分为手术组(n=10)及假手术组(n=10),手术组行RDN术,假手术组仅行肾动脉造影。3、监测实验犬体重、血压,术后6月无痛处死实验犬,取心脏组织,行Masson染色观察心肌纤维化程度,计算胶原容积分数(CVF)、周围血管胶原面积比率(PVCA), Real-time PCR检测组织miR-133a表达水平;Real-time PCR、Western-blot检测组织CTGF表达。结果:1、建模期假手术组、手术组实验犬体重、收缩压(SBP)、舒张压(DBP)、平均动脉压(MAP)均明显升高,与基线、对照组相比均有统计学差异(P<0.05),假手术组、手术组组间对比无统计学差异(P>0.05);2、经导管射频消融去...
