酯酰辅酶A:胆固醇酰基转移酶1抑制鼻咽癌增殖和转移的研究

研究背景:鼻咽癌是一种来源于鼻咽部的恶性肿瘤,具有明显的地域、种族和民族分布。鼻咽癌在我国的南方以及东南亚地区发病率较高,男、女性发病率分别为30/10万(人/年)和10/10万(人/年)。目前研究表明鼻咽癌的病因是遗传易感性、EB病毒(epstein-barr virus,EBV)感染、环境因素和饮食致癌物的相互作用,其发病是多步骤、多因素且复杂的过程。由于鼻咽癌初期无明显症状,早期即出现淋巴转移,大多数确诊的鼻咽癌患者多处于II-IVb期。目前,鼻咽癌的治疗方案比较有限,常规治疗是以顺铂为基础的化疗联合调强放疗(IMRT)。尽管目前的治疗手段已经显著提高了鼻咽癌患者的5年生存率,但仍有约20%的患者因远处转移和局部复发而导致治疗失败。因此,探索鼻咽癌转移的机制,寻找新的抑制肿瘤转移的治疗靶点,是目前鼻咽癌基础研究的热点之一。能量代谢异常是肿瘤细胞十大特征之一,它为肿瘤细胞提供了生存优势。酮体作为体内重要的代谢产物,与肿瘤的发生和演进密切相关。正常神经元和神经胶质细胞可以有效的代谢酮体并通过酮体代谢产生能量,而包括胶质瘤在内的多种肿瘤细胞不能利用酮体作为能量底物,而是作为脂质合成的前...
