氟化钠对雌性大鼠生殖功能的影响

【摘要】:目的 氟化钠对人类生殖功能的影响,尤其是对雌性生殖的影响是越来越受到研究者们的关注。然而有关氟化钠对雌性生殖影响的研究报道很是少见,影响的机制尚不清楚。本研究旨在探索氟化钠对雌性生殖的影响极其相关机制。 方法 构建SD大鼠氟化钠中毒模型。将SD雌性大鼠随机分为三组,即对照组,100mg/L组和200mg/L组;分别饮用蒸馏水,浓度为100mg/L和200mg/L的氟化钠溶液,染毒180天。染毒期结束后各组雌性大鼠分别与雄性大鼠交配观察其妊娠率变化。采用ELISA法检测氟化钠染毒大鼠血清生殖激素浓度(E2,P, FSH, LH和T)的改变。用H-E染色的方法观察各组雌性大鼠子宫组织形态学变化。采用免疫组织化的方法对子宫雌激素α受体和孕激素受体进行组织学定位。利用Western blot方法检测雌激素α受体和孕激素受体在不同浓度的氟化钠染毒SD雌性大鼠子宫中的表达情况。 结果 1、氟化钠染毒组大鼠妊娠率显著降低。 2、血清中E2、P、FSH、LH和T浓度分别显著低于对照组,染毒组雌性大鼠子宫显微结构发生了明显异常改变。 3、氟化钠暴露组大鼠子宫雌激素α受体和孕激素受体表达较对照组明显上调。 结论 氟化钠对雌性生殖功能造成了显著影响,,降低了雌性大鼠妊娠率。生殖激素的降低,雌激素受体及孕激素受体蛋白的异常表达可能是氟化钠对雌性生殖功能影响的机制之一。
[Abstract]:Purpose. The effects of sodium fluoride on human reproductive function, especially on female reproduction, have attracted increasing attention from researchers. However, studies on the effects of sodium fluoride on female reproduction are rare. The effect of sodium fluoride on female reproduction is not clear. Method. The SD female rats were randomly divided into three groups: the control group 100 mg / L and the 200 mg / L group, drinking distilled water, 100 mg / L and 200 mg / L sodium fluoride solution, respectively. At the end of the exposure period, female rats in each group were mated with male rats to observe their pregnancy rate. Serum reproductive hormone concentrations of E2P, FSH, LH and T4 were detected by ELISA method. Methods the morphologic changes of uterus in each group of female rats were observed. The histological localization of estrogen 伪 receptor and progesterone receptor in uterus was performed by immunohistochemical method. The estrogen 伪 receptor and pregnancy excitation were detected by Western blot method. Expression of hormone receptor in uterus of SD female rats exposed to different concentrations of sodium fluoride. Results. 1. The pregnancy rate of rats exposed to sodium fluoride decreased significantly. (2) the serum levels of FSHLH LH and T in serum were significantly lower than those in the control group, and the microstructures of the uterus in the exposed group were significantly different from those in the control group. 3. The expression of estrogen 伪 receptor and progesterone receptor in uterus of rats exposed to sodium fluoride was significantly higher than that of control group. Conclusion. Sodium fluoride had a significant effect on female reproductive function and decreased the pregnancy rate of female rats. The abnormal expression of estrogen receptor and progesterone receptor protein may be one of the mechanisms of sodium fluoride affecting female reproductive function.
