饮食热能限制对小鼠学习记忆及脑内IRP-2和CREB基因表达的影响

【摘要】:目的以脑内铁调节蛋白-2(iron regulated protein-2,IRP-2)和c AMP反应元件结合蛋白(c AMP response element binding protein,CREB)基因表达为观测点,探讨饮食热能限制(caloric restriction,CR)对小鼠学习记忆能力的影响。方法雄性小鼠40只随机均分为正常对照组(normal control group,NC group)、CR组、脑老化组(brain aging group,BA group)、CR+BA组,每组10只,Morris水迷宫试验测定学习空间记忆能力,以逆转录聚合酶链反应法(reverse transcription-polymerase chain reaction,RT-PCR)检测小鼠大脑IRP-2及CREB基因的表达水平。结果与NC组相比,BA组逃避潜伏期(escape latency,EL)显著增高(P0.05);CR+BA组的EL显著低于BA组(P0.05),而与NC组无显著差异(P0.05);CR组与NC组的EL相比无明显差异(P0.05)。BA组IRP-2、CREB表达水平显著低于NC组(P0.05);CR组IRP-2、CREB表达水平与NC组相比无明显差异(P0.05);CR+BA组IRP-2、CREB表达水平显著高于BA组(P0.05),与CR组及NC组相比无明显差异(P0.05)。结论 CR可以上调BA组小鼠脑内IRP-2及CREB基因表达,提示CR能够通过影响上述两种基因的表达而增强BA组小鼠的学习记忆能力。NC小鼠给予CR后,其脑内IRP-2及CREB基因的表达无明显变化。
[Abstract]:Objective to observe the expression of iron-regulated protein -2iron regulated protein-2IRP-2 and c AMP response element binding protein in the brain. To investigate the effect of caloric restriction on the learning and memory ability of mice, 40 male mice were randomly divided into normal control group group group control group, brain aging group group BA group group and CR BA group. The ability of learning spatial memory was measured by Morris water maze test in 10 rats in each group. The expression of IRP-2 and CREB genes in the brain of mice was detected by reverse transcription-polymerase chain reaction- transcription-polymerase RT-PCR.Results compared with NC group, escape latencies of escape latencies in BALB group were significantly higher than those in control group, and the El of CR BA group was significantly lower than that in BA group, but had no significant difference with NC group. There was no significant difference in the expression of IRP-2CREB between the CR group and the NC group. The expression of IRP-2CREB in the P0.05- BA group was significantly lower than that in the NC group. There was no significant difference in the expression of IRP-2CREB between the CR group and the NC group. The expression level of IRP-2CREB in the CR group was significantly higher than that in the control group, and in comparison with the CR group and the NC group, the expression level of IRP-2CREB in the CR group was significantly higher than that in the control group. The expression of IRP-2CREB in the CR group was significantly higher than that in the NC group. Conclusion CR can up-regulate the expression of IRP-2 and CREB genes in the brain of BA group mice. These results suggest that CR can enhance the learning and memory ability of BA group mice by affecting the expression of these two genes. After CR, the expression of IRP-2 and CREB genes in the brain of the mice treated with CR has no obvious change.
