镧对仔鼠海马核转录因子-κB表达影响

【摘要】:目的探讨亚慢性氯化镧(La Cl3)暴露对子代大鼠海马组织核转录因子-κB(NF-κB)表达的影响。方法健康成年雌性Wistar大鼠24只,随机分为对照组及0.25%、0.5%、1.0%La Cl3组,染镧组仔鼠自雌鼠受孕至断乳后1个月通过自由饮水摄入镧;利用免疫组化法检测海马p-IKKα/β的分布和表达;应用Western blot法检测海马组织p-IκBα和p-NF-κBP65的表达。结果 0.25%、0.5%、1.0%La Cl3组仔鼠海马CA1区p-IKKα/β表达分别为(54.72±10.04)、(40.73±4.90)、(32.99±5.42),CA3区表达分别为(64.80±8.27)、(47.78±4.92)、(36.17±7.71),DG区表达分别为(303.67±34.78)、(166.83±29.8)、(87.01±17.40);CA1、CA3、DG区各染镧组仔鼠海马3区的p-IKKα/β表达与对照组比较,除DG区0.25%La Cl3组外其他各染镧组仔鼠海马p-IKKα/β表达均低于对照组(P0.05);0.25%、0.5%、1.0%La Cl3组仔鼠海马p-IκBα和p-NF-κBp65蛋白表达IDV值分别为(0.58±0.06)和(0.46±0.06)、(0.49±0.07)和(0.36±0.08)、(0.36±0.09)和(0.25±0.08),均低于对照组的(0.68±0.05)和(0.56±0.08)(P0.05),仔鼠海马p-IκBα和p-NF-κBp65蛋白表达均与La Cl3染毒剂量呈负相关(r=-0.994、-0.978,P0.05)。结论亚慢性La C13染毒可引起子代大鼠海马细胞NF-κB活性降低,可能是镧引起学习记忆能力下降的机制之一。
[Abstract]:Objective to investigate the effect of subchronic lanthanum chloride (La Cl 3) exposure on the expression of nuclear transcription factor 魏 B (NF- 魏 B) NF- 魏 B in hippocampus of offspring rats. Methods Twenty-four healthy adult female Wistar rats were randomly divided into two groups: the control group and the 0.25g / 0.50g / 1.0La Cl3 group. In lanthanum group, lanthanum was ingested through free drinking water one month after weaning, and the distribution and expression of p-IKK 伪 / 尾 in hippocampus were detected by immunohistochemical method. Western blot method was used to detect the expression of p-I 魏 B 伪 and p-NF- 魏 BP65 in hippocampal tissue. Results the expression of p-IKK 伪 / 尾 in the CA1 area of hippocampus of 0.25g / g group was 54.72 卤10.04 卤10.04 and 32.99 卤5.42CA3, respectively. The expression of p-IKK 伪 / 尾 in the CA1 area of rats in the 0.2550 卤4.92La Cl3 group was respectively 303.67 卤34.786.83 卤29.87.87.01 卤40CA1CA3DG and 64.80 卤8.2777.78 卤4.92DG, respectively, compared with the control group, and the expression of IKK / 尾 in the hippocampus of the control group was higher than that of the control group, and the expression of p-IKK 伪 / 尾 in the hippocampus of the control group was significantly higher than that of the control group (P < 0.05), and the expression of p-IKK 伪 / 尾 in the hippocampal CA1 area of the rats in the group of 0.25 卤0.567 卤34.786.83 卤29.87.01 卤17.01 卤17.01 卤17.01 卤17.01 卤17.01 卤17.01 卤17.01, respectively. The expression of p-IKK 伪 / 尾 in hippocampus of rats in lanthanum group except DG area 0.25La Cl3 group was lower than that in control group (P 0.05 + 0.05) and 0.56 卤0.056 卤0.056 卤0.05in La Cl3 group respectively. The expression of p-I 魏 B 伪 and p-NF- 魏 Bp65 in hippocampus were 0.58 卤0.06, 0.46 卤0.06 卤0.07) and 0.36 卤0.036 卤0.036 卤0.08 in La Cl3 group, respectively, which were lower than those in control group (0.68 卤0.05) and 0.56 卤0.08P0.05A, respectively. The expression of p-I 魏 B 伪 and p-NF#en4# 魏 魏 in hippocampus of neonatal rats were lower than those in control group (0.68 卤0.05) and 0.56 卤0.08P0.05A, respectively. There was a negative correlation between the expression of white and the dose of La Cl3. Conclusion the activity of NF- 魏 B in hippocampal cells of filial generation rats can be decreased by subchronic La C13 exposure. It may be one of the mechanisms of the decline of learning and memory induced by lanthanum.
