三类非离子表面活性剂对细胞增殖抑制作用的研究

【摘要】:本文采用噻唑蓝(MTT)比色法评价了4种烷基酚聚氧乙烯醚系列表面活性剂(APEO)对人宫颈癌细胞(HeLa)及中华仓鼠卵巢细胞(CHO)的细胞毒性;采用MTT比色法评价了3种N,N-二烷基脲系列化合物对人胚皮肤成纤维细胞(CCC-ESF-1)的增殖抑制作用;采用3H-TdR渗入法评价了3种脂肪醇聚氧乙烯醚类表面活性剂(AEO)对人胚皮肤成纤维细胞的增殖抑制作用。较为系统地研究了三类物质的细胞毒性。实验结果表明:三类物质对细胞的增殖抑制率均随其浓度的增大而增大,但毒性差异明显。烷基酚聚氧乙烯醚OP-7、OP-9、NP-10和NP-25的浓度为0.1mg/ml时,其对HeLa细胞的增殖抑制率分别为90.23%、87.76%、89.56%和24.86%;对CHO细胞的增殖抑率分别为89.34%、85.95%、90.97%和21.31%。AEO-5、AEO-9和AEO-15浓度为0.1mg/ml时,对皮肤细胞有较强的增殖抑制作用,抑制率分别为99.53%、98.60%和63.15%。而乙基脲、丙基脲和丁基脲浓度为0.1mg/ml时,对皮肤细胞的增殖抑制作用不明显,当浓度为5mg/ml时,对皮肤细胞具有了较强的增殖抑制作用,抑制率分别为61.94%、62.50%和86.94%。因此可知,这三类物质达到一定浓度后,均对细胞的生长有明显的抑制作用,并呈现浓度依赖性,但N,N-二烷基脲对细胞的毒性远小于烷基酚聚氧乙烯醚和脂肪醇聚氧乙烯醚;脂肪醇聚氧乙烯醚和烷基酚聚氧乙烯醚类化合物EO数增大,对细胞的毒性逐渐减小;三种N,N-二烷基脲类物质随碳链长度的增大,对细胞的毒性也逐渐增大。
[Abstract]:The cytotoxicity of four kinds of alkylphenol polyoxyethylene ether (APEO) on human cervical cancer cell line (HeLa) and Chinese hamster ovarian cell (Cho) was evaluated by MTT colorimetry. MTT colorimetric assay was used to evaluate the inhibitory effects of three N- N- dialkylurea series on the proliferation of human embryonic skin fibroblasts (CCC-ESF-1). The inhibitory effects of three fatty alcohol polyoxyethylene ether surfactants (AEO) on the proliferation of human embryonic skin fibroblasts were evaluated by 3H-TdR method. The cytotoxicity of three kinds of substances was systematically studied. The inhibition rate of cell proliferation increased with the increase of its concentration. When the concentration of OP-7 and NP-25 was 0.1 mg / ml, the inhibition rates of the proliferation of HeLa cells were 90.23% and 89.56%, respectively, and the inhibition rates of CHO cells were 89.34% and 21.31%, respectively. AEO-5AEO-9 and AEO-15 concentration were 0.1 mg / ml, respectively, in the presence of alkylphenol polyoxyethylene ether (OP-7OP-9NP-10) and NP-25 (0.1 mg / ml), the inhibitory rates on the proliferation of CHO cells were 87.76% and 24.86%, respectively, when the concentrations of OP-7 and OP-9NP-10 and NP-25 were 0.1 mg / ml, respectively, the inhibitory rates on the proliferation of CHO cells were 89.34 45%, 21.31% and 21.31%, respectively. The inhibition rate was 98.60% and 63.15%, respectively. When the concentration of ethyl urea, propyl urea and Ding Ji urea was 0.1 mg / ml, the inhibitory effect on the proliferation of skin cells was not obvious, and when the concentration was 5 mg / ml, the inhibitory effect on the proliferation of skin cells was not obvious. The inhibition rates were 61.94%, 62.50% and 86.94%, respectively. Therefore, when these three kinds of substances reached a certain concentration, they all had obvious inhibitory effects on the growth of the cells, and showed a concentration-dependent manner. However, the toxicity of N-dialkylurea to cells was much less than that of alkylphenol polyoxyethylene ether and fatty alcohol polyoxyethylene ether, and the EO number of fatty alcohol polyoxyethylene ether and alkylphenol polyoxyethylene ether compounds increased, and the toxicity of fatty alcohol polyoxyethylene ether and alkyl phenol polyoxyethylene ether compounds decreased gradually. The cytotoxicity of three N- N-dialkyl ureas increased with the length of carbon chain.
