百草枯对雄性大鼠精液质量及睾丸组织脂质过氧化的影响

【摘要】:[目的]研究百草枯(paraquat,PQ)对雄性大鼠精液质量和睾丸组织脂质过氧化的影响。[方法]将32只健康成年雄性Wistar大鼠随机分为4组,即对照组和低、中、高三个PQ染毒组(0.5、2.0、8.0 mg/kg),每组8只。采用经口灌胃,连续染毒8周。染毒结束后,处死大鼠,分离睾丸及附睾组织,称重并计算脏器系数,分析精子数量及活力,采用H-E染色观察睾丸组织病理学变化,并检测睾丸组织中丙二醛(MDA)含量、超氧化物岐化酶(SOD)以及谷胱甘肽过氧化物酶(GSH-Px)活力。[结果]低、中、高染毒组大鼠精子数量分别为(48.37±3.76)×10~6、(41.56±3.13)×10~6、(32.48±4.09)×10~6,活力分别为(68.85±5.27)%、(66.65±2.83)%、(52.30±1.99)%,均低于对照组[(53.19±4.15)×10~6,(70.65±6.07)%],差异有统计学意义(P0.05)。各染毒组大鼠睾丸脏器系数(0.87±0.08、0.85±0.07、0.78±0.07)及附睾脏器系数(0.27±0.02、0.29±0.03、0.31±0.03)与对照组(0.94±0.06,0.24±0.03)相比较,差异亦具有统计学意义(P0.05)。与对照组相比,染毒组睾丸组织出现明显的退行性改变;且高PQ染毒组睾丸组织中MDA含量升高[(1.45±0.18)nmol/mg vs.(0.79±0.11)nmol/mg],SOD[(76.19±6.71)μ/mg vs.(108.50±11.24)μ/mg]及GSH-Px[(15.07±2.71)μ/mg vs.(30.61±3.23)μ/mg]活力下降。[结论]百草枯导致大鼠精液质量下降,睾丸组织脂质过氧化增强。
[Abstract]:[objective] to study the effects of paraquat PQ) on semen quality and lipid peroxidation in testis of male rats. [methods] Thirty-two healthy adult male Wistar rats were randomly divided into four groups: control group, low and middle group. Eight rats in each group were given oral oral administration for 8 weeks. The rats were killed, testicular and epididymal tissues were isolated, weighed, and organ coefficients were calculated, and sperm count and motility were analyzed. The histopathological changes of testis were observed by H-E staining, and the contents of malondialdehyde (MDA), the activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) in testis were detected. The difference was also statistically significant (P 0.05). Compared with the control group, the testicular tissue of the exposed group showed obvious degenerative changes. The activity of MDA in testis was increased [1.45 卤0.18nmol / mg vs.(0.79 卤0.11nmol / mg] and GSH-Px (15.07 卤2.71 渭 / mg vs.(30.61 卤3.23 渭 / mg) decreased. [conclusion] Paraquat induced the decrease of semen quality and the increase of lipid peroxidation in testis.
