【摘要】:以"红元帅"苹果的干燥叶子为试材,通过单因素试验确定苹果叶总黄酮的最佳提取工艺条件,采用可见分光光度法对苹果叶中总黄酮含量进行测定,以期为苹果叶中总黄酮提取工艺及含量测定提供方法和依据。结果表明:苹果叶总黄酮的最佳提取工艺条件为乙醇作溶剂,乙醇浓度60%,料液比1∶20g/mL,提取温度为70℃,提取时间为30min;在选定试验条件下对苹果叶进行含量测定,对芦丁对照品浓度与吸光度进行线性回归,其回归方程为Y=24.821 43X-0.031,R2=0.999 45(n=6),芦丁对照品质量在4~24μg范围内呈良好的线性关系,在此条件下测定苹果叶中总黄酮平均含量为30.28mg/g。试验优选出了苹果叶中总黄酮的最佳提取工艺条件及含量测定方法,该方法简单、快速,准确,精密度,稳定性,重现性良好。
