白藜芦醇对肥胖妊娠SD大鼠胚胎存活及后代抗氧化能力的研究

【摘要】:本研究旨在考察白藜芦醇(resveratrol, RES)对肥胖妊娠SD大鼠胚胎存活的影响,采用Diquat为应激源研究母体灌服RES对后代抗氧化能力的影响。试验包括以下两部分: 试验一白藜芦醇对肥胖妊娠SD大鼠胚胎存活和抗氧化能力影响 本试验选用40只SPF级雌性SD大鼠随机分为2个处理组:高脂日粮组(20%脂肪)和标准日粮组(7%脂肪),每个处理20个重复,每个重复1只SD大鼠。SD大鼠适应1周后,供给处理日粮,饲喂8周后成功构建营养性肥胖模型(高脂日粮组雌鼠体重量为标准日粮组雌鼠体重量的20%,记为实验性肥胖大鼠),随后大鼠雌:雄=1:1合笼配种,检查阴户发现阴栓,则记为妊娠当天E(0)。在此基础上,选取正常妊娠鼠和肥胖妊娠鼠各20只,采用2×2因子,设计四个处理组,分别是高脂日粮组、高脂日粮灌服白藜芦醇(200mg/kg.bw)组、标准日粮组、标准日粮灌服白藜芦醇(200mg/kg.bw)组。每个处理妊娠大鼠各10只,每个重复1只,妊娠第3天至17天灌服RES,于妊娠第18天每个处理分别选取4只妊娠大鼠屠宰取样,考察RES对肥胖妊娠SD大鼠繁殖成绩及抗氧化能力的影响。结果表明:1)试验各组胎儿重和胎盘重差异不显著(P0.05),但高脂口粮灌服RES显著提高了母体妊娠第18天的胚胎存活率(p0.05)。2)与高脂日粮相比,高脂日粮灌服RES组甘油三酯(TG)浓度降低53%(p0.05)、游离脂肪酸(FFA)降低28%(P0.05)。3)与高脂日粮组相比,高脂日粮灌服RES能显著提高母体肝脏、胎盘、胎肝的总抗氧化能力(T-AOC)和谷胱甘肽过氧化物酶(GSH-Px)活性(P0.05),降低丙二醛(MDA)浓度(P0.05),但对母体肝脏、胎盘、胎肝的总超氧化物歧化酶(T-SOD)及过氧化氢酶(CAT)活性没有显著影响。 试验二妊娠期母鼠白藜芦醇摄入对后代抗氧化能力的影响 本试验采用2×2×2因子设计,分别选用试验一四个处理组剩余的6只妊娠大鼠的分娩子鼠,直至哺乳子鼠22天断奶,断奶后代分别按高脂日粮、高脂日粮灌服白藜芦醇、标准日粮、标准日粮灌服白藜芦醇进行分组,每个处理8个重复,每个重复1只,子鼠均饲喂同一标准日粮,以Diquat为应激源,从第23天开始,每间隔一天上午8:00腹腔注射Diquat (8mg/kg.BW,用0.9%的灭菌生理盐水稀释),共7次,对照组注射相同剂量的灭菌生理盐水。断奶后第14天早晨眼眶静脉采血,分离血清,测定其抗氧化指标。结果表明:腹腔注射Diquat应激源后,与高脂日粮组相比,高脂日粮灌服白藜芦醇组的子鼠断奶后第7天和14天体重分别增加12%和25%,血清T-AOC和CAT分别提高33%和37%(P0.05),MDA浓度降低16%(P0.05),标准日粮灌服白藜芦醇组子鼠断奶后第7天和14天体重分别增加1%和3%,血清T-AOC和CAT提高1.5%和1%,MDA浓度降低3%。 上述结果表明,RES具有降低肥胖妊娠SD大鼠体重的趋势,显著改善母体、胎盘和胎儿抗氧化能力,提高胚胎存活率;同时母体妊娠期灌服RES可提高后代抗氧化能力。
[Abstract]:The purpose of this study was to investigate the effects of resveratrol resveratrolle (res) on the embryo survival of obese SD rats. Diquat was used as the stress source to study the effect of maternal administration of RES on the antioxidant capacity of offspring.The experiment consists of the following two parts:Effects of resveratrol on embryo survival and antioxidant capacity in obese SD rats with pregnancyIn this experiment, 40 SPF female SD rats were randomly divided into two groups: high fat diet group (20% fat) and standard diet group (7% fat).After feeding for 8 weeks, a nutritional obesity model was successfully established (the weight of female rats in the high-fat diet group was 20% of that in the standard diet group, recorded as experimental obese rats, and then the female: male 1: 1 cage was bred, and the vulva was found to have negative embolism.It was recorded as the day of pregnancy.On this basis, 20 normal pregnant rats and 20 obese pregnant rats were selected, and 2 脳 2 factors were used to design four treatment groups: high fat diet group, high fat diet fed with resveratrol 200 mg / kg 路bww group, standard diet group.The standard diet was fed with resveratrol 200 mg / kg 路bw.Ten pregnant rats were treated with RES3 to 17 days of gestation, and 4 pregnant rats were slaughtered on the 18th day of pregnancy.To investigate the effect of RES on reproductive performance and anti-oxidation ability of obese pregnant SD rats.The results showed that there was no significant difference in fetal weight and placental weight among the three groups, but the embryo survival rate on the 18th day of gestation was significantly increased by RES administration with high fat ration compared with the high fat diet.Compared with the high fat diet group, the concentration of triglyceride (TG) and free fatty acid (FFA) in the high fat diet group were decreased by 53% and 28% (P 0.05) 路3 respectively. Compared with the high fat diet group, the high fat diet administration of RES could significantly improve the maternal liver and placenta.The total antioxidant capacity (T-AOC) and glutathione peroxidase (GSH-Px) activity of fetal liver (T- AOC) and glutathione peroxidase (GSH-Px) activity (P0. 05) decreased the concentration of malondialdehyde (MDA) (P0. 05), but had no significant effect on the activities of total superoxide dismutase (T-SOD) and catalase (CAT) in maternal liver, placenta and fetal liver.Effect of resveratrol intake on antioxidant capacity of offspring in pregnant ratsIn this experiment, 2 脳 2 脳 2 factor design was used to test the remaining 6 pregnant rats in four treatment groups, which were weaned until 22 days after weaning. After weaning, they were fed with high fat diet, high fat diet, resveratrol, standard diet, respectively.Resveratrol was administered with standard diet, 8 replicates in each treatment, and 1 repeats in each treatment. The offspring were fed with the same standard diet. Diquat was used as the stress source from the 23rd day.Diquat 8 mg / kg 路BW was injected intraperitoneally at 8:00 every day and diluted with 0.9% sterilized saline for 7 times. The control group was injected with the same dose of sterilized saline.On the 14th day after weaning, blood was collected from orbital vein, serum was isolated and its antioxidant index was measured.The results showed that: after intraperitoneal injection of Diquat stress source, compared with high-fat diet group,In resveratrol group, the body weight increased by 12% and 25% on the 7th and 14th day after weaning, the serum T-AOC and CAT increased by 33% and 37%, respectively. The concentration of T-AOC decreased by 16% and decreased by 16%. The rats fed with standard diet on the 7th and 14th day after weaning were given resveratrol.The body weight increased by 1% and 3%, the serum T-AOC and CAT increased by 1.5% and the concentration of MDA decreased by 3%.The results indicated that res had the tendency to decrease the body weight of obese SD rats, improve the antioxidation ability of mother, placenta and fetus, and increase the survival rate of embryos, and the maternal administration of RES during pregnancy could improve the antioxidant ability of offspring.
