二氧化硫体内衍生物对大鼠卵巢功能毒性的初步研究

【摘要】:研究背景:随着工业化的不断发展和城市的不断扩张,大气污染已日趋严重,威胁着人类赖以生存的环境和全球的可持续发展性。二氧化硫(SO_2)是主要的大气污染物之一。石油、煤、天然气的燃烧以及工业上各种有色金属的冶炼、石油精制、硫化橡胶及造纸等均可产生SO_2,挥发于空气中对大气造成严重污染。SO_2作为一种有毒性气体,可造成脑、肺、心、肝等脏器受到损伤,严重时可影响其功能,危机生命;人体若1天内摄取4-6g,会引起剧烈的胃肠功能障碍,造成腹泻。二氧化硫可引起全身毒性反应,二氧化硫吸入体内后会行成衍生物—亚硫酸钠:亚硫酸氢钠以物质的量之比为3:1存在于体内,随着血液分布全身。 目的:研究二氧化硫体内衍生物对大鼠性激素水平的影响以及对卵巢的毒性作用。初步探讨二氧化硫体内衍生物对卵巢的毒性作用,提高人们对二氧化硫毒性认识,严格控制环境中及生活中二氧化硫浓度,增强全人类环保意识。 方法:选择健康的60天左右,体重(200g 20)g清洁级成年雌性未孕WISTER大鼠,(山西医科大学动物实验中心);每日给予标准光照,颗粒饲料喂养,自由摄食,室温20-25℃相对湿度50%-65%。每日晨9:00至11:00取阴道分泌物涂片,巴氏染色,光镜观察性周期,选取30只性周期规律的大鼠进入实验。将30只大鼠随机分为对照组、低剂量组、高剂量组,对照组采用生理盐水1ml,每天腹腔注射一次,,连续14天;低剂量组采用二氧化硫体内衍生物混合液1ml,浓度0.5g/kg,每天腹腔注射一次,连续14天;高剂量组采用混合液1ml,浓度1g/kg,每天腹腔注射一次,连续14天;给药结束后于动情期,尾静脉采血,用放射免疫法检测卵泡刺激素(FSH)、血清雌二醇(E2),黄体生成素(LH)激素水平的变化。股动脉放血处死大鼠,取卵巢组织固定、切片、HE染色,观察卵巢组织形态学变化。 结果:(1)低剂量组与高剂量组均有中毒症状,以高剂量组为著,雌性大鼠可出现饮食、活动减少,于腹腔注射后可出现震颤、肌肉痉挛等中毒症状。(2)三个组之间比较,FSH有显著差异(F=84.19,P㩳0.0001),SO_2各剂量组中大鼠FSH较对照组显著增加;且高剂量组的影响显著高于低剂量组(P㩳0.05)。SO_2对大鼠血清中LH水平的影响显著(F=57.92,P㩳0.0001);高剂量组中LH水平较低剂量组、对照组显著增大(P㩳0.05);低剂量组和对照组没有显著差异。三个组之间比较,E2有显著差异(F=153.83,P㩳0.0001),染毒组各剂量组中的大鼠血清中E2水平较对照组有显著降低;高剂量组的影响显著高于低剂量组(P㩳0.05)。(3)高剂量组大鼠卵巢组织的病理改变主要表现为始基卵泡和初级卵泡增多,而次级卵泡和成熟卵泡较少见,且闭锁卵泡增多。结论:二氧化硫体内衍生物可引起雌性大鼠性轴功能紊乱及卵巢组织学改变。
[Abstract]:Background: with the development of industrialization and the expansion of cities, air pollution is becoming more and more serious, which threatens the environment on which human beings depend and the sustainable development of the whole world.So _ 2 is one of the main atmospheric pollutants.The combustion of petroleum, coal and natural gas, as well as the smelting of various non-ferrous metals in industry, petroleum refining, vulcanized rubber and paper making can all produce so _ 2, which volatilizes in the air and causes serious air pollution. So _ 2 as a toxic gas, it can cause brain and lung.Heart, liver and other organs are damaged, when serious can affect its function, crisis life; if the human body to ingest 4-6g in one day, will cause severe gastrointestinal dysfunction, causing diarrhea.Sulfur dioxide can cause systemic toxic reactions. After inhalation of sulfur dioxide, sodium sulfite: sodium bisulfite exists in the body with the weight ratio of 3:1, along with the distribution of blood throughout the body.Aim: to study the effects of sulfur dioxide derivatives on sex hormone levels and ovarian toxicity in rats.To explore the toxic effect of sulfur dioxide derivatives on ovary, to improve people's understanding of sulfur dioxide toxicity, to strictly control the concentration of sulfur dioxide in the environment and in daily life, and to enhance the awareness of environmental protection of all mankind.Methods: healthy adult female WISTER rats with a body weight of 200g 20g for about 60 days were selected (Animal Experimental Center of Shanxi Medical University).
