芦丁对三甲基锡损害学习记忆功能的保护作用与拮抗突触囊泡蛋白表达下调有关

【摘要】:目的探讨芦丁对三甲基锡(Trimethyltin,TMT)损害小鼠学习记忆功能的保护作用及可能机制。方法以6~9周龄雄性BALB/c小鼠为研究对象,随机分为生理盐水组(对照)、TMT组、TMT+芦丁组、芦丁组,每组各10只;建立TMT(2.25 mg/kg·B.W.)急性暴露模型,后两组芦丁(10 mg/kg·B.W.)预处理1周,均腹腔注射;TMT给药后24 h,Morris水迷宫测试各组的逃逸潜伏期,Western检测各组海马和皮层脑区突触囊泡蛋白(Synaptophysin,SYP)的表达情况。结果 Morris水迷宫显示TMT给药后24 h,与TMT组相比,对照组和芦丁组及TMT+芦丁组逃逸潜伏期显著缩短(P0.05),与芦丁组及对照组相比,TMT+芦丁组逃逸潜伏期无统计学意义(P0.05);Western显示TMT给药后24 h,与对照组相比,TMT组海马和皮层脑区SYP蛋白表达显著降低(P0.05);与TMT组相比,TMT+芦丁组海马和皮层脑区SYP蛋白表达显著升高(P0.05),与芦丁组及对照组相比,TMT+芦丁组海马和皮层脑区SYP蛋白表达无统计学意义(P0.05)。结论芦丁预处理对TMT急性暴露损害小鼠学习记忆功能有保护作用,其保护作用可能与拮抗海马和皮层脑区SYP表达下调有关。
[Abstract]:Objective to investigate the protective effect of rutin on learning and memory impairment in mice and its possible mechanism. Methods male BALB/c mice aged 6 to 9 weeks were randomly divided into normal saline group (control group, 10 rats in each group), and TMT(2.25 mg/kg B.W. Acute exposure model, the latter two groups of rutin 10 mg/kg B.W. After preconditioning for 1 week, the escape latency and the expression of synaptic vesicle protein (Synaptophysin sinusii SYP) in hippocampus and cortical brain were detected by Morris water maze test 24 hours after intraperitoneal injection of TMT. Results Morris water maze showed that 24 hours after administration of TMT, compared with TMT group. The escape latency of control group, rutin group and TMT rutin group was significantly shortened than that of rutin group and control group. There was no significant difference in escape latency between rutin group and rutin group. Western analysis showed that the escape latency of rutin group was 24 hours after administration of TMT, and that of hippocampus and cortical brain in TMT group was higher than that in control group. Compared with TMT group, the expression of SYP protein in hippocampus and cortical brain was significantly increased in SYP group, and no significant difference was found in SYP protein expression in hippocampus and cortical brain in Rutin group compared with rutin group and control group. Conclusion Rutin preconditioning has protective effect on learning and memory function in mice with acute exposure to TMT, which may be related to the antagonism of down-regulation of SYP expression in hippocampus and cortical brain.
