1,3-丁二烯作业工人遗传损伤与细胞周期蛋白D1基因多态性的关系

【摘要】:目的探讨1,3-丁二烯(BD)作业工人外周血淋巴细胞遗传物质损伤状况及其与细胞周期蛋白D1(CCND1)基因多态性的关系。方法采用外周血淋巴细胞胞质分裂阻滞微核(CBMN)试验和彗星试验评价171名BD作业工人和55名非职业暴露对照人群的染色体和DNA损伤水平,应用聚合酶链反应-限制性片段长度多态性方法分析CCND1基因的多态性。同时,通过问卷调查的方式了解被调查者的年龄、性别、吸烟、饮酒和个人职业暴露史。结果 BD作业工人外周血淋巴细胞微核率和彗星尾距分别为(7.17±5.09)‰和4.65(95%CI 3.49~5.98),均高于对照组(3.82±2.37)‰和2.38(95%CI 0.82~3.98),差异有统计学意义(P0.01);暴露组中具有CCND1基因A870G位点GG基因型个体的彗星尾距显著低于AA基因型个体(P0.05)。结论在现有BD暴露水平下,外周血淋巴细胞胞质分裂阻滞微核法和彗星试验均能检出BD作业工人的遗传物质损伤;CCND1等位基因A870G位点GG基因型可能与职业性BD暴露导致的外周血淋巴细胞DNA损伤水平增加风险相关联。
[Abstract]:Objective to investigate the genetic material damage in peripheral blood lymphocytes of workers exposed to 1-butadiene-butadiene (BDB) and its association with the polymorphism of cyclin D _ (1) C _ (CND _ 1) gene. Methods the chromosomal and DNA damage levels in 171 BD exposed workers and 55 non-occupational exposed controls were evaluated by cytoplasmic mitotic block micronucleus (DNA) assay and comet assay in peripheral blood lymphocytes. Polymerase chain reaction-restriction fragment length polymorphism (RFLP) was used to analyze the polymorphism of CCND1 gene. At the same time, the age, sex, smoking, drinking and personal occupational exposure were investigated by questionnaire. Results the micronucleus rate and comet tail distance of peripheral blood lymphocytes in BD exposed workers were 7.17 卤5.09 鈥,
