牛磺酸对染锰大鼠丘脑氨基酸类神经递质及其相关酶改变的干预作用

【摘要】:目的:探讨牛磺酸对染锰大鼠丘脑氨基酸类神经递质及其相关酶改变的干预作用。 方法:按随机方法将SD大鼠分为空白对照组、染锰组、预防组、治疗组和治疗对照组,共5组。染锰组、预防组、治疗组和治疗对照组大鼠腹腔注射MnCl2-4H2O20mg/kg·d,其中预防组大鼠同时腹腔注射牛磺酸200mg/kg·d,空白对照组腹腔注射等容量生理盐水,连续8周,每周5天,染锰组、预防组和空白对照组试验终止。然后,治疗组大鼠继续腹腔注射牛磺酸200mg/kg·d,治疗对照组大鼠腹腔注射等容量生理盐水,连续8周。最后一次注射后24h进行水迷宫实验,分别以前5天平均逃避潜伏期及低6天的平台搜索次数评估大鼠空间学习能力及空间记忆能力的变化;然后处死大鼠,切取丘脑。用BCA蛋白浓度测定试剂盒测定样品中的蛋白含量,用高效液相色谱(]IPLC)紫外检测法测定大鼠脑丘脑谷氨酸(glutamic acid, Glu)、谷氨酰胺(glutamine, Gln)和Y-氨基丁酸(gamma-aminobutyric acid, GABA)的含量;用酶联免疫法(ELISA)测定谷氨酸脱羧酶(GAD)、磷酸激活谷氨酰胺酶(PAG)和谷氨酰胺合成酶(GS)的含量。 结果:1、染锰组大鼠逃避潜伏期时间较空白对照组明显延长(P0.05),搜索到目标区域次数明显减少,差异有统计学意义(P0.05)。预防组大鼠逃避潜伏时间较染锰组明显减少(P0.05),预防组大鼠搜索目标区域次数与染锰组比较其差异有统计学意义(P0.05)。治疗组大鼠逃避潜伏期较治疗对照组明显缩短(P0.05),治疗组大鼠搜索目标区域次数与治疗对照组比较其差异有统计学意义(P0.05)。治疗组大鼠逃避潜伏时间较染锰组明显减少(P0.05),与空白对照组比较其差异有统计学意义(P0.05);治疗组大鼠搜索目标区域次数与染锰组比较其差异有统计学意义(P0.05)2、染锰8周后,染锰组Glu含量比空白对照组含量高,差别有统计学意义(P0.05);染锰组G1n和GABA含量比空白对照组低,其差异有统计学意义(P0.05);染锰组PAG含量比空白对照组含量高,差异有统计学意义(P0.05),染锰组GAD和GS含量比空白对照组低,差异有统计学意义(P0.05);牛磺酸预防8周时,预防组Glu含量比染锰组含量低,差异有统计学意义(P0.05);预防组G1n和GABA含量比染锰组高,其差异有统计学意义(P0.05);预防组PAG含量比染锰组含量低,差异有统计学意义(P0.05),预防组GAD和GS含量比染锰组高,差异有统计学意义(P0.05)。牛磺酸治疗8周后,治疗组Glu含量均比治疗对照组含量低,治疗组G1n和GABA含量均比治疗对照组含量高,差异有统计学意义(P0.05);治疗组PAG含量比治疗对照组含量低,治疗组GAD和GS含量均比治疗对照组含量高,差异有统计学意义(P0.05)。治疗组Glu含量比染锰组含量低,差异有统计学意义(P0.05);治疗组G1n和GABA含量比染锰组高,其差异有统计学意义(P0.05);治疗组PAG含量比染锰组含量低,差异有统计学意义(P0.05),治疗组GAD和GS含量比染锰组高,差异有统计学意义(P0.05)。 结论:锰可致大鼠丘脑Glu升高,Gln和GABA减低,牛磺酸对锰所致大鼠丘脑Glu、Gln和GABA的改变有干预作用。
[Abstract]:Objective : To investigate the effect of taurine on amino acid neurotransmitters and their associated enzymes in rats with manganese - stained rats .


Methods : Sprague - Dawley rats were randomly divided into two groups : blank control group , manganese - stained group , prevention group , treatment group and control group .
The contents of glutamic acid ( Glu ) , glutamine ( glutamine , Gln ) and gamma - aminobutyric acid ( GABA ) were determined by high performance liquid chromatography ( HPLC ) UV detection method .
The contents of glutamate decarboxylase ( GAD ) , phosphoactivated glutamine ( PAG ) and glutamine synthetase ( GS ) were determined by enzyme linked immunosorbent assay ( ELISA ) .


Results : 1 . Compared with the control group , the number of escape latency in the treatment group was significantly shorter than that in the control group ( P0.05 ) . The number of escape latency in the treatment group was significantly shorter than that in the control group ( P0.05 ) .
Compared with the control group , the content of Glu in the treated group was higher than that of blank control group ( P0.05 ) .
The content of G1n and GABA in the exposed group was lower than that in the blank control group ( P0.05 ) .
The content of PAG in the manganese - stained group was higher than that in the blank control group ( P0.05 ) . The GAD and GS contents of the manganese - stained group were lower than those in the blank control group ( P0.05 ) .
When taurine was used for 8 weeks , the content of Glu in the prevention group was lower than that in the exposed group ( P0.05 ) .
The content of G1n and GABA in the prevention group was higher than that in the manganese - stained group ( P0.05 ) .
Compared with the control group , the content of Glu in the treatment group was lower than that in the control group ( P0.05 ) , and the content of G1n and GABA in the treatment group was higher than that in the control group ( P0.05 ) .
The content of PAG in the treatment group was lower than that in the control group , the content of GAD and GS in the treatment group was higher than that in the control group ( P0.05 ) . The Glu content in the treatment group was lower than that in the control group ( P0.05 ) .
The content of G1n and GABA in the treatment group was higher than that in the manganese - stained group ( P0.05 ) . The content of PAG in the treatment group was lower than that of the manganese - stained group ( P0.05 ) . The GAD and GS contents in the treatment group were higher than those in the manganese - stained group ( P0.05 ) .


Conclusion : The changes of Glu , Gln and GABA in the hypothalamus of rats induced by manganese can increase , Gln and GABA decrease , taurine has an interference effect on the changes of Glu , Gln and GABA in the thalamus induced by manganese .
