新疆双峰驼乳清蛋白组分对人宫颈癌HeLa细胞增殖的抑制作用

【摘要】:目的:探讨新疆双峰驼乳清蛋白组分对人宫颈癌HeLa细胞增殖的抑制作用.方法:采用四甲基偶氮唑盐(MTT)法测定新疆双峰驼乳蛋白组分对HeLa细胞增殖的抑制作用;荧光染色检测HeLa细胞凋亡的形态变化;倒置显微镜、扫描电镜和透射电镜观察HeLa细胞的形态变化;用流式细胞术检测HeLa细胞凋亡率、线粒体膜电位的变化.结果表明:新疆双峰驼乳清蛋白组分TR35可抑制人宫颈癌HeLa细胞的增殖并诱导其凋亡,并呈时间和剂量依赖性(P0.01);倒置显微镜、扫描电镜和透射电镜观察到典型的凋亡特征,荧光显微观察到凋亡小体;Annexin-Ⅴ/PI双染检测出现早期凋亡细胞;线粒体膜电位去极化,并将细胞周期阻滞在S期.结论:新疆双峰驼乳清蛋白组分TR35可抑制HeLa细胞的增殖和诱导细胞凋亡,TR35组分诱导细胞凋亡的途径可能与线粒体通路有关.
[Abstract]:Aim: to investigate the inhibitory effect of whey protein on the proliferation of human cervical cancer HeLa cells in Xinjiang. Methods: the inhibitory effect of lactoprotein on the proliferation of HeLa cells was determined by tetramethylazolium tetrazolium (MTT) method. The morphological changes of apoptosis of HeLa cells were detected by fluorescence staining. The morphological changes of HeLa cells were observed by scanning electron microscope and transmission electron microscope, and the apoptosis rate and mitochondrial membrane potential of HeLa cells were detected by flow cytometry. The results showed that the whey protein component TR35 could inhibit the proliferation and induce apoptosis of human cervical cancer HeLa cells in a time-and dose-dependent manner, and the typical apoptotic characteristics were observed by inverted microscope, scanning electron microscope and transmission electron microscope.
