磷酸三苯酯对青春期雄性SD大鼠睾丸增殖细胞核抗原和芳香化酶表达的影响

【摘要】:目的: 观察磷酸三苯酯对青春期大鼠睾丸组织的影响,及其对相关基因和蛋白表达的影响,初步研究磷酸三苯酯对大鼠生殖系统的作用。 方法: 将40只健康的23日龄SPF级雄性SD大鼠随机分为4组,每组10只,即溶剂对照组,低剂量组(250mg/kg)、中剂量组(500mg/kg)、高剂量组(750mg/kg);配成相应的玉米油溶液,按0.5%体重灌胃。连续灌胃染毒30天后,取大鼠双侧睾丸组织,运用HE染色观察各实验组大鼠睾丸组织的病理改变;免疫组织化学染色观察各组大鼠睾丸PCNA和AROM蛋白的表达;应用RT-PCR检测各组大鼠睾丸PCNA、 AROM mRNA的表达:应用Western blot检测各组大鼠睾丸PCNA、 AROM蛋白的表达。 结果: 与对照组相比,染毒组大鼠的体重明显下降(P0.05),而四个组相应的睾丸脏器系数无统计学意义(P0.05)。HE染色显示:对照组大鼠的生精小管结构正常,各级生精细胞排列有序。各染毒组可见生精上皮发生变性,各级生精细胞排列紊乱,甚至出现了精子扭曲缠结,肿胀,脱落的现象(P0.05)。免疫组化检测结果显示,PCNA、AROM在染毒剂量组睾丸组织的阳性表达强度降低,与对照组比较其差别有显著性(P0.05)。RT-PCR结果表明,各剂量组大鼠睾丸组织的PCNA、 AROM mRNA表达降低,与对照组相比具有统计学意义(P0.05)。Western blot结果显示,各剂量组大鼠睾丸的PCNA、AROM蛋白表达明显降低,与对照组相比具有统计学意义(P0.05)。 结论: TPP能抑制大鼠睾丸PCNA、AROM mRNA和蛋白的表达,提示雄性大鼠青春期暴露于TPP,可以通过损伤睾丸组织结构,降低PCNA、 AROM表达水平等机制,从而对大鼠的睾丸发育产生损害。
[Abstract]:Objective: To observe the effect of triphenyl phosphate on testis of pubertal rats, and to study the effect of triphenyl phosphate on the reproductive system of rats. Methods: Forty healthy 23-day-old male SD rats of SPF grade were randomly divided into 4 groups, 10 rats in each group: solvent control group, low dose group (250 mg / kg), middle dose group (500 mg / kg), high dose group (750 mg / kg), and corresponding corn oil solution (0.5% body weight). After 30 days of intragastric administration, the testis of the rats were collected and the pathological changes of testis in each experimental group were observed by HE staining, and the expressions of PCNA and AROM protein in testis of each group were observed by immunohistochemical staining. RT-PCR was used to detect the expression of PCNA, AROM mRNA in testis of rats and Western blot was used to detect the expression of PCNA and AROM in testis. Results: Compared with the control group, the body weight of the rats in the exposed group decreased significantly (P 0.05A), while the viscera coefficients of the testis in the four groups were not statistically significant (P 0.05). He staining showed that the structure of the seminiferous tubules in the control group was normal, and the spermatogenic cells of the four groups were arranged in an orderly manner. The spermatogenic epithelium was denatured, the spermatogenic cells of all levels were disordered, and the spermatozoa were twisted and tangled, swelled and shedded (P0.05). The results of immunohistochemistry showed that the positive expression intensity of PCNA, AROM mRNA in testicular tissue of the exposed group was lower than that of the control group. The results of RT-PCR showed that the expression of PCNA, AROM mRNA in the testis of each dose group was lower than that of the control group. Compared with the control group, the results of blot showed that the expression of PCNAA-AROM in testis of each dose group was significantly lower than that of the control group (P 0.05), and that of the control group was significantly lower than that of the control group (P 0.05). Conclusion: TPP can inhibit the expression of PCNAA-AROM mRNA and protein in testis of rats, suggesting that the mechanism of male rats exposed to TPP during puberty may damage the testis development by damaging the structure of testis and decreasing the expression level of AROM and so on.
