沙海蜇胶原蛋白肽对光老化小鼠皮肤的保护作用及体外透皮吸收研究

【摘要】:1本文以沙海蜇皮为研究对象,讨论了腌渍沙海蜇皮的基本组成,从其基本组成可知腌渍沙海蜇皮中最主要的成分是水分和盐分,在干物质中最主要的成分是蛋白质,其中蛋白质中胶原蛋白约占到三分之一,腌渍沙海蜇中高水分、高盐分,较低的胶原蛋白的纯度为其中的胶原蛋白的提取增加了难度,需要寻找一种有效的提取方式对沙海蜇中的胶原蛋白进行大规模的提取以进行后续实验。因此本文讨论了提取胶原蛋白常用的三种方法(酶法提取、热水提取和酸法提取)所得三种胶原蛋白PSC(酶溶性蛋白),HEG(热水提取胶原蛋白),ASC(酸溶性胶原蛋白)的理化性质。结果表明三种方法得到的胶原蛋白结构相似。酸法提取可以得到纯度较高的胶原蛋白,其羟脯氨酸含量高于PSC和HEG,并且ASC具有良好的乳化稳定性。HEG分子量相对较大,且具有良好的乳化性。其吸湿性由强到弱依次为PSCHEGASC,保湿性最好的样品为PSC。综合各种因素的考量,本实验最终选用热水对胶原蛋白进行大规模的提取。并将所得的胶原蛋白进一步的酶解,超滤成不同分子量的肽段。 2以紫外诱导光老化的小鼠皮肤为研究对象,研究了海蜇胶原蛋白对其皮肤的保护作用。实验小鼠共分为11组,即正常对照组,模型对照组,阳性对照组,胶原蛋白高剂量组以及胶原蛋白低剂量组,Mr8000高、低剂量组,2500Mr8000高、低剂量组,Mr2500高、低剂量组。通过对小鼠皮肤与血清中生化指标的检测和组织形态的观察可以得到如下结论:沙海蜇胶原蛋白能够抑制紫外照射引起的胶原蛋白和水份的流失,其中Mr8000对抑制紫外光照射小鼠皮肤中胶原蛋白的流失最有效而2500Mr8000的胶原肽可以较好的抑制皮肤中水分的流失;沙海蛰胶原肽可以通过提高抗氧化物酶SOD、GSH-Px、CAT的活力起到抗氧化的作用,其中Mr2500的胶原肽对皮肤组织中抗氧化物酶活力的提高起到较好的效果;沙蛰胶原蛋白肽可以有效的改善紫外照射小鼠皮肤的组织形态,使其接近正常小鼠皮肤的组织形态;综上所述,沙海蜇胶原蛋白对光老化小鼠的皮肤具有保护作用。 3从分子量、剂量、时间、pH值等几个方面沙海蜇胶原蛋白肽的透皮吸收性能进行研究。从研究中我们发现分子量越小的胶原蛋白多肽表现出越好的透皮吸收性能。并且通过累计透皮吸收量和时间的曲线得到了沙海蜇胶原蛋白肽体外透皮吸收的零级动力学方程,, Mr2500: Q=209.29t-6.801;2500Mr8000:Q=84.381t+133.75;Mr8000: Q=52.597t+72.96。随着样品浓度的升高,沙海蜇胶原蛋白肽的体外透皮吸收性能也随之提高。在pH为8左右时,沙海蜇胶原蛋白多肽表现出较好的体外透皮性能。
[Abstract]:1 the basic composition of salted jellyfish was discussed in this paper. From its basic composition, the water and salt were the main components in pickled jellyfish, and protein was the most important component in dry matter. The protein of collagen accounts for about 1/3, the pickled jellyfish has high water content and high salinity, and the purity of collagen increases the difficulty of extracting collagen. We need to find an effective way to extract collagen from jellyfish. Therefore, the physicochemical properties of three common methods of extracting collagen (enzymatic extraction, hot water extraction and acid extraction) were discussed in this paper. The results showed that the collagen obtained by the three methods was similar in structure. The content of hydroxyproline in collagen extracted by acid method was higher than that of PSC and HEG.The ASC had good emulsifying stability and its molecular weight was relatively large and had good emulsifying property. The order of hygroscopicity from strong to weak is PSCHEGASC.The sample with the best moisture retention is PSC. Considering various factors, hot water was used to extract collagen on a large scale. The collagen was further hydrolyzed and ultrafiltration into peptides with different molecular weight. 2 the protective effect of collagen of jellyfish on the skin of mice skin induced by ultraviolet irradiation was studied. The experimental mice were divided into 11 groups: normal control group, model control group, positive control group, collagen high dose group and collagen low dose group. The results showed that the collagen of jellyfish could inhibit the loss of collagen and water caused by ultraviolet irradiation. Mr8000 is the most effective method to inhibit the loss of collagen in the skin of mice irradiated by ultraviolet light, and the collagen peptide of 2500Mr8000 can effectively inhibit the loss of water in the skin. The collagen peptide of Mr2500 can improve the activity of antioxidant enzyme GSH-PxCAT, and the collagen peptide of Mr2500 can improve the activity of antioxidant enzyme in skin tissue. Jellyfish collagen peptide can effectively improve the skin tissue morphology of ultraviolet irradiated mice, make it close to the normal mouse skin tissue morphology; in summary, jellyfish collagen has a protective effect on the skin of photoaged mice. 3The transdermal absorption of collagen peptide of jellyfish was studied from molecular weight, dose, time and pH value. We found that collagen peptides with lower molecular weight showed better transdermal absorption. The zero-order kinetic equation of collagen peptide transdermal absorption of jellyfish in vitro was obtained by the curve of cumulative transdermal absorption amount and time. Mr 2500: QN 209.29t-6.801 / 2500 Mr 8000: QN 84.381t 133.75m8000: QN 52.597t 72.96. With the increase of sample concentration, the in vitro transdermal absorption of collagen peptide from jellyfish was improved. When pH was about 8, the collagen peptide of jellyfish showed better transdermal properties in vitro.
