芹菜素对雄性大鼠生殖系统功能的影响及相关机制探讨

【摘要】:目的研究芹菜素对雄性大鼠生殖系统功能的影响并探讨相关机制,为芹菜素的进一步开发利用提供科学依据。方法48只SD成年雄性大鼠随机分为对照组(生理盐水10ml/kg-bw)和芹菜素低剂量组(234 mg/kg-bw)、中剂量组(468 mg/kg-bw)、高剂量组(936mg/kg·bw)。1次/天,6天/周,连续5周给予动物生理盐水或芹菜素灌胃。(1)处死动物,分离睾丸、附睾、精囊腺、前列腺、尿道球腺、肛提肌/球海绵体肌称重,计算脏器系数。(2)游离附睾中的精子,采用WLJY-9000系统分析精子密度、运动参数。制备精子伊红染色涂片,镜下观察精子形态,计算精子畸形率。(3)制备睾丸HE染色病理切片和电镜切片,观察睾丸组织结构和细胞超微结构的变化。(4)制备睾丸单细胞悬液,流式细胞仪检测睾丸细胞周期。(5)制备睾丸组织匀浆,检测睾丸氧化还原指标T-AOC、SOD、CAT、GSH-Px、GSH、MDA及睾丸组织ACP、AKP、LDH、SDH、NOS、NADPH和Na+K+-ATP酶、Ca2+Mg2+-ATP酶。(6)在灌胃的第7、14、21、28、35天活体心内取血,测定血清T、E2、ICSH、FSH水平。结果(1)各剂量组的睾丸、附睾、阴茎、精囊腺、前列腺、尿道球腺、肛提肌/球海绵体肌的湿重和脏器系数与对照组比较,差异无统计学意义(P0.05)。(2)高剂量组a级精子密度低于对照组,d级精子密度高于对照组(P0.01);高剂量组精子运动参数VCL. VSL低于对照组,差异有统计学意义(P0.01)。高剂量组精子畸形率高于对照组,差异有统计学意义(P0.05),畸形类型主要为尾折叠。(3)低、中、高剂量组睾丸组织结构呈不同程度的损伤:生精小管结构不规则,间质变薄;生精上皮变薄,细胞层数减少,细胞排列混乱,管腔内成熟精子数目减少;长形精子细胞出现于生精上皮各层细胞中间。电镜下观察到,高剂量组精子尾部线粒体肿胀、线粒体嵴变形。(4)各剂量组睾丸G0/G1期、S期和G2/M期细胞与对照组比较,差异无统计学意义(P0.05)。(5)低、中剂量组T-AOC活力高于对照组,高剂量组T-AOC活力低于对照组(P0.05),低剂量组MDA含量低于对照组(P0.05),低剂量组SOD活力、中剂量组GSH-Px活力低于对照组(P0.05),低、中、高剂量组GSH含量均低于对照组(P0.05)。低、中、高剂量组ACP、AKP和LDH活力均低于对照组(P0.01),低、中、高剂量组Ca2+Mg2+-A-TP酶活力和高剂量组Na+K+-ATP酶活力高于对照组(P0.05)。(6)中、高剂量组T浓度在14天时高于对照组,差异有统计学意义(P0.05);高剂量组E2浓度在21天时低于对照组,差异有统计学意义(P0.05);中剂量组T/E2在28天时高于对照组,高剂量组T/E2在14天和21天时高于对照组(P0.05)。低、高剂量组在28天时,血清ICSH浓度高于对照组(P0.05);各剂量组血清FSH浓度在各时间点与对照组比较,差异无统计学意义(P0.05)。结论(1)936 mg/kg·bw AP降低大鼠精子质量,降低精子运动速度,升高精子畸形率,损伤睾丸组织结构和精子尾部线粒体超微结构。(2)AP对睾丸组织的氧化损伤和能量代谢的干扰可能是精子质量下降的原因。(3)AP可引起血清性激素水平变化,其机制及其对精子质量的影响还需进一步研究。
[Abstract]:Objective To study the effects of apigenin on the reproductive system function of male rats and to explore the related mechanism to provide scientific basis for the further exploitation and utilization of apigenin . Methods 48 SD adult male rats were randomly divided into control group ( normal saline 10ml / kg - bw ) and apigenin low - dose group ( 234 mg / kg - bw ) . Results ( 1 ) The levels of serum T , E2 , ICSH and FSH were significantly lower than those in control group ( P0.05 ) .
High dose group sperm motility parameter VCL . There was significant difference in sperm deformity rate in high - dose group than in control group ( P0.05 ) . The difference was statistically significant ( P0.05 ) .
The number of mature sperms was decreased and the number of mature sperms in the lumen was decreased .
The activity of GSH - Px in the low - dose and medium - dose groups was lower than that in the control group ( P0.05 ) . The activity of the low - dose group and the high - dose group were lower than those in the control group ( P0.05 ) .
The concentration of E2 in high dose group was lower than that of control group at 21 days ( P0.05 ) .
The concentration of T / E2 in the middle dose group was higher than that of the control group at 28 days , and the T / E2 in the high dose group was higher than that of the control group ( P0.05 ) at the 14th day and the 21st day . The serum ICSH concentration was higher in the high dose group than the control group ( P0.05 ) .
Conclusion ( 1 ) 936 mg / kg 路 bw AP can decrease sperm quality in rats , decrease sperm motility , increase sperm deformity rate , damage testis structure and ultrastructure of sperm tail mitochondria .
