3种食用菌分级多糖消脂护肝功能研究

【摘要】:通过热水提取、乙醇沉淀的方式获得榆黄菇、毛木耳和灰树花多糖,利用分子量截留的超滤方法获得15个分级多糖组分。采用酒精和脂肪酸(FFA)共同诱导Hep G2细胞建立脂肪肝模型,以细胞存活率和胞内甘油三酯含量为检测指标评价分级多糖消脂护肝作用。研究结果发现PS V、AP V和GF V 3个分子量高于100 k Da的组分是各自品种分级多糖中的优势组分,比例分别为71.06%、34%和68.89%;3种分级多糖可以提高肝损伤细胞的存活率分别为27.61%、37.68%和29.25%,同时减少甘油三酯在细胞内的堆积,具有明显的消脂护肝作用。
[Abstract]:Polysaccharides of Ulmus officinalis, Auricularia pubescens and Grifola frondosa were obtained by hot water extraction and ethanol precipitation, and 15 fractionated polysaccharides were obtained by ultrafiltration with molecular weight interception. The fatty liver model of HepG2 cells was induced by alcohol and fatty acid FFA.The survival rate and intracellular triglyceride content of HepG2 cells were used to evaluate the effect of fractionated polysaccharides on lipids and liver protection. The results showed that the three components with molecular weight of more than 100kDa were the dominant components in the classification of polysaccharides. The ratio of 71.06% and 68.89% were increased by 27.61% and 29.25%, respectively, and triglyceride accumulation in the cells was reduced, which had obvious antilipidemic and protective effects on the liver. The results showed that the lipopolysaccharide could increase the survival rate of the injured liver cells by 27.61% and 29.25% respectively, while reducing the accumulation of triglycerides in the cells.
