纳米氧化铜对家兔的急性眼刺激性

【摘要】:目的观察纳米氧化铜(Cu O)对家兔急性眼刺激性作用以及及时冲洗措施对于缓解急性刺激症状的有效性。方法40、80和200 nm近球形纳米Cu O颗粒用于试验。纳米Cu O先以0.5%羟丙基甲基纤维素(HPMC)分散后以生理盐水稀释配制成染毒分散悬液。新西兰家兔用于试验。试验设40、80和200 nm纳米Cu O组(分别按照10、40和160 mg/L剂量分组)和0.5%HPMC组(分散剂对照),每组3只动物。试验按照染毒不冲洗和染毒后4 s冲洗两种方案进行。不冲洗试验:将纳米Cu O测试液0.1 ml分别滴入到3只家兔左眼结膜囊中,并使眼睛被动闭合1 s,右眼不处理作自身对照;4 s冲洗试验:家兔染毒后第4秒,用无菌生理盐水彻底冲洗眼部30 s。并于染毒后1、24、48、72 h、4和7 d对动物眼睛进行检查和评分。结果 40 nm粒径三个剂量组和80 nm粒径40和160 mg/L剂量组动物左眼于染毒后(不冲洗)1 h出现眼结膜明显充血,至24 h出现进行性加重。80 nm和40 nm纳米Cu O染毒组动物分别于染毒后48和72 h眼结膜充血时症状明显缓解,至观察第4天,所有动物充血症状消散。上述动物角膜和虹膜未观察到损伤改变。200 nm粒径纳米Cu O所有剂量组动物均未观察到眼部刺激症状。根据急性眼刺激性试验结果显示,40 nm和80 nm粒径纳米Cu O眼刺激性反应分级为可逆性眼损伤。在4 s冲洗试验中,40、80和200 nm组所有动物均未出现明显眼部刺激症状。试验中0.5%HPMC对所有动物不具有眼刺激性。结论小粒经的40和80 nm纳米Cu O具有结膜损伤眼刺激性。及时(4 s)采取眼部冲洗措施能有效降低纳米Cu O接触引起的急性眼刺激症状。
[Abstract]:Objective to observe the effect of nano-cupric oxide (CuO) on acute eye irritation in rabbits and the effectiveness of timely washing measures in relieving acute irritation symptoms. Methods 40,80 nm and 200 nm spherical nano-CuO particles were used in the experiment. Nano CuO was first dispersed with 0.5% hydroxypropyl methyl cellulose (HPMC) and then diluted with normal saline to form the dispersive suspension. New Zealand rabbits are used in experiments. The rats were divided into 4080 nm and 200 nm nano-CuO groups (10 40 and 160 mg / L respectively) and 0.5 HPMC (dispersant control group, 3 animals in each group). The experiment was carried out according to the two schemes of no washing after exposure and washing at 4 s after exposure. No washing test: 0.1 ml nano-CuO test solution was dripped into the conjunctival sac of 3 rabbits, and the eyes were passively closed for 1 s, and the right eye was not treated for 4 s as a self-control flushing test: the rabbits were treated 4 seconds after exposure. The eyes were washed thoroughly with sterile saline for 30 s. The eyes of the animals were examined and scored on the 4th and 7th day after exposure. Results the conjunctiva hyperemia was observed in the 40 nm diameter three dose groups and the 80 nm 40 and 160 mg / L groups after exposure (1 h without washing). The symptoms of conjunctival hyperemia were alleviated at 48 and 72 hours after 24 h of progressive exacerbation at 80 nm and 40 nm nano-CuO, respectively. At the 4th day of observation, the hyperemia symptoms of all animals dissipated. No damage changes were observed in the cornea and iris of the above animals. No ocular irritation was observed in all dose groups. According to the results of acute eye irritation test, the irritation reaction of 40 nm and 80 nm nanocrystalline CuO eyes was classified as reversible eye injury. In the 4 s irrigation test, no obvious ocular irritation symptoms were found in all the animals in the 4080 and 200 nm groups. In the experiment, 0.5 HPMC was not eye irritating to all animals. Conclusion 40 nm and 80 nm nanocrystalline CuO particles have ocular irritation in conjunctival injury. Eye washing can effectively reduce the symptoms of acute eye irritation caused by nano Cu O contact.
