50Hz磁场暴露后大鼠生物学变化的临床检验分析

【摘要】:目的随着经济的发展和信息时代的来临,各种类型的电子产品及电子设备在家庭和工作环境中的大量使用,使人们暴露于50Hz电磁场的机会与日俱增。电器产品给人们带来了诸多方便的同时,其电磁辐射等负面效应逐步引起人们的广泛关注。本研究以临床常用血液检验和病理检测技术为手段,系统观察50Hz磁场三种等比剂量暴露对大鼠血液学和生物化学指标和相关脏器组织形态学的影响规律,旨在进一步研究观察50Hz磁场的生物学效应,,尝试寻找相关血清标志物,为接触者健康监护和卫生事业管理(包括特殊健康检查项目的确定、健康监护档案管理、健康状况分析、评价预防措施效果、修订卫生标准等)提供线索和依据。 方法将SD雄性大鼠随机分为1个对照组和3个辐照组,分别全身暴露于0μT(对照组)、20μT、100μT及500μT50Hz磁场中,每天2小时,连续4周,每周测其体重变化,在暴露结束后采集外周血和心、肝、肾、睾丸等组织,检测血清肝功能、肾功能、血脂、心肌酶谱、睾酮水平和外周血细胞数量等指标,称重并计算心、肝、肾、睾丸的脏器系数,光镜病理学分析各脏器组织形态结构的变化。 结果50Hz磁场暴露期间各组大鼠体重无明显差异。20μT和100μT剂量暴露对血清肝功能、肾功能、血脂、心肌酶谱、睾酮水平、各组织脏器系数及外周血细胞数量均无明显影响。500μT剂量暴露后大鼠血清肌酐水平较对照组显著升高(P0.05),总胆固醇水平较对照组明显降低(P0.05),血清睾酮水平较对照组显著升高(P0.05),心、肝、肾、睾丸组织脏器系数均无明显差异(P0.05),外周血白细胞数量较对照组显著增加(P0.05)。进一步相关分析发现,血清肌酐、总胆固醇、睾酮、外周血白细胞数量与50Hz磁场暴露水平之间存在明显的相关关系(p均0.05);血清葡萄糖和肌酸激酶水平与50Hz磁场暴露水平之间均存在正相关关系趋势(p均0.05)。HE染色发现,大鼠心肌间质血管扩张充血;肝中央静脉及肝窦扩张充血,周围肝细胞水肿;肾小管上皮细胞浊肿,间质血管扩张充血;睾丸间质组织可见局灶性充血水肿现象;以上各组织形态结构的变化随着暴露剂量的增加,充血水肿越明显,呈一定的剂量-效应关系。 结论在本实验条件下,50Hz磁场暴露对雄性大鼠生长发育无明显影响,可引起脏器组织呈现一定的血管通透性改变;高剂量(500μT)暴露可引起雄性大鼠肾脏功能、睾丸功能、血脂代谢以及外周血白细胞数量的变化;血清肌酐、总胆固醇、睾酮、外周血白细胞数量可能是50Hz磁场暴露生物学效应潜在的血清标志物,需要进一步研究证实。
[Abstract]:Objective with the development of economy and the advent of the information age, various types of electronic products and electronic devices are widely used in the home and work environment, which make people exposed to 50Hz electromagnetic field day by day. Electrical products bring people a lot of convenience, at the same time, its electromagnetic radiation and other negative effects gradually aroused widespread concern. In this study, the effects of 50 Hz magnetic field on hematological and biochemical indexes and histomorphology of related organs in rats were systematically observed by means of blood test and pathological examination. The aim of this study was to investigate the biological effects of 50Hz magnetic field and to search for serum markers for health monitoring and health management (including the determination of special health examination items, health monitoring file management, health status analysis). To evaluate the effect of preventive measures, to revise the health standards, etc.) to provide clues and evidence. Methods Sprague-Dawley male rats were randomly divided into one control group and three irradiation groups. The rats were exposed to 0 渭 T 100 渭 T 100 渭 T and 500 渭 T 50Hz magnetic field for 2 hours a day for 4 weeks. The body weight was measured weekly. The peripheral blood and heart were collected after exposure. Liver, kidney, testis, serum liver function, renal function, blood lipid, myocardial enzyme spectrum, testosterone level and the number of peripheral blood cells were measured, and the organ coefficients of heart, liver, kidney and testis were calculated. Histological changes of various organs were analyzed by light microscopy. Results during 50 Hz magnetic field exposure, there was no significant difference in body weight between each group. The dose of 20 渭 T and 100 渭 T had no effect on serum liver function, renal function, blood lipid, myocardial enzyme spectrum and testosterone level. The serum creatinine level, total cholesterol level and serum testosterone level were significantly higher than those in the control group (P0.05), and the serum testosterone level was significantly higher than that of the control group (P0.05), and the serum testosterone level was significantly higher than that of the control group (P0.05), and the total cholesterol level was significantly lower than that of the control group (P0.05), and the serum testosterone level was significantly higher than the control group (P0.05). There was no significant difference in organ coefficient among liver, kidney and testis (P0.05), and the number of peripheral blood leukocytes was significantly increased compared with control group (P0.05). Further correlation analysis showed that there was a significant correlation between serum creatinine, total cholesterol, testosterone, the number of peripheral white blood cells and 50 Hz magnetic field exposure (p0.05). There was a positive correlation between serum glucose and creatine kinase levels and 50 Hz magnetic field exposure (p0.05) .HE staining showed that the myocardial interstitial vasodilation of rats was congested, the central hepatic vein and sinusoidal dilatation hyperemia, and the peripheral hepatocyte edema. Renal tubule epithelial cell turbid, interstitial vascular dilatation and hyperemia; testicular interstitial tissue showed focal hyperemia and edema. The changes of morphology and structure of the above tissues increased with the increase of exposure dose, the hyperemia and edema became more obvious. There is a dose-effect relationship. Conclusion under this condition, 50 Hz magnetic field exposure has no obvious effect on the growth and development of male rats, and can cause certain vascular permeability changes in visceral tissues, and high dose (500 渭 T) exposure can induce renal function and testicular function in male rats. Serum creatinine, total cholesterol, testosterone and the number of peripheral white blood leukocytes may be potential serum markers for biological effects exposed to 50 Hz magnetic field.
