低剂量X线照射对内皮祖细胞功能影响的研究

【摘要】:目的:本研究拟通过探讨低剂量X线照射(LDI)对大鼠骨髓来源的内皮祖细胞功能的影响。 方法:取SD大鼠(体重200g)股骨、胫骨,反复冲洗骨髓腔,离心后取单核细胞细,接种于25cm2的培养瓶中,置于37℃、体积分数为0.05的二氧化碳、饱和湿度的培养箱中培养。使用流式细胞仪鉴定细胞后,实验组给予25mGy、50mGy、75mGy、100mGy低剂量X线及1Gy高剂量照射,对照组则不予照射。照射后分别于6h、1d、3d、5d、7d天计算细胞数及CCK-8检测细胞增殖情况,照射后分别在第1d、3d、5d行Western-blot检测细胞表面标记:VEGFR-2;RT-PCR法测VEGFR-2mRNA的表达;免疫组织化学染色法测定细胞VWF、VEGFR-2的表达。 结果:低剂量照射后实验组较对照组的细胞计数有明显增强;大剂量组可抑制细胞的增殖。CCK-8法检测结果表明低剂量照射后,可明显刺激内皮祖细胞的增殖,与对照组相比有统计学意义(P0.05)。而1Gy高剂量照射则会抑制内皮祖细胞增殖(P0.05)。实验组VEGFR-2在1d、3d、5d的表达高于对照组。而且随着剂量的增加,表达量随之升高(P0.05)。试验组VEGFR-2mRNA的表达也显著强于对照组(P0.05)。免疫组织化学显示在1和3天,实验组处的VWF和VEGFR-2表达强于对照组(P0.05)。 结论:低剂量X线照射后对内皮祖细胞有兴奋效应,表现为细胞生长加速,同时在一定时间内血管形成相关基因表达增强。
