丙烯酰胺对雌性Wistar大鼠学习记忆和长时程增强的影响及其可能机制

【摘要】:目的研究重复sc给予丙烯酰胺(AM)28 d后对学习记忆的影响及其可能的作用机制,以及单次sc给予AM后对大鼠海马高频刺激后群峰电位(PS)幅度改变的影响。方法雌性Wistar大鼠分别sc给予AM 10,20和40 mg·kg-1(给药当天计为第1天),连续给予28 d。每周记录1次体质量。于第22~第27天进行Morris水迷宫实验,评价AM对大鼠空间定位能力的影响;分别于第29和第30天进行跳台实验,观察5 min内大鼠跳下平台的次数以及首次跳下平台的时间。取AM 40 mg·kg-1组大鼠海马组织,Western蛋白印迹法检测N-甲基-D-天冬氨酸受体(NR)2A及2B与Ⅱ型钙调蛋白激酶(Ca MKⅡ)的表达与磷酸化水平。另取Wistar雌性大鼠,单次sc给予AM 40 mg·kg-1,进行长时程增强(LTP)实验,观察高频刺激后PS的改变。结果与正常对照组相比,给予AM后,大鼠体质量增长减缓(P0.01)。AM 20及40 mg·kg-1组大鼠找到平台的时间显著延长,但是对大鼠穿越平台的次数无显著影响。与实验首日(第29天)相比,经过电击强化学习记忆,正常对照组大鼠为躲避电击,次日(第30天)5 min内跳下平台的次数显著减少(P0.01),且首次跳下平台的时间明显延长(P0.01),而AM 10,20和40 mg·kg-1组5 min内的错误次数以及首次跳下平台的时间无显著改变。Western蛋白印迹结果显示,与正常对照组相比,AM 40 mg·kg-1组大鼠海马组织中NR2A及NR2B的表达与磷酸化水平显著增加,且Ca MKⅡ磷酸化水平显著升高(P0.01)。LTP结果显示,与正常对照组相比,单次sc给予AM 40 mg·kg-1显著降低Wistar雌性大鼠海马的PS幅度(P0.01),损伤了大鼠海马部位的突触可塑性。结论 AM可能通过改变谷氨酸的含量,增加NR的表达与活性,导致钙超载,引发神经细胞死亡,从而使LTP的PS幅度下降,影响海马神经突触的可塑性,最终影响中枢的学习记忆功能。
