饮用水有机提取物对正常人肝细胞凋亡及炎性因子的影响
[Abstract]:Objective to observe the changes of interleukin 6 (interleukin 6), alanine aminotransferase (alanine transaminase,ALT) and apoptosis induced by organic extracts from drinking water in L02 cells. Methods solid phase extraction was used to extract organic pollutants from drinking water. The normal human liver (L02) cells at logarithmic growth stage were exposed to the medium containing the final concentration of 0.312 ~ 0.625 ~ 1.2500 ~ 2.500 ~ 5.000 L/ml for 24 ~ 4872 h, respectively. The cells were exposed to a solvent control (including 0.1%DMSO medium) and a blank control (culture medium) respectively. Apoptosis rate, IL-6 content and ALT activity in supernatant of cell culture medium were measured. Results compared with the solvent control group, the apoptosis rate of L02 cells was higher at different time after exposure to organic extracts from drinking water at different time points, except for the groups exposed for 24 h and 0.312 5 L/ml, respectively. The content of IL-6 in the supernatant of L02 cells was higher at different time after exposure to organic extracts from drinking water (P0.05). The activity of ALT in the supernatant of L02 cells exposed to different doses of drinking water organic extract at different time was higher than that of the control group (P 0.05), except for the treatment group of 0.312 ~ 0.625 L/ml for 24 h (P0.05). The apoptosis rate, IL-6 content and ALT activity in the supernatant of L02 cells were increased with the increase of drinking water organic extract dose and the prolongation of exposure time. Conclusion in this experiment, drinking water organic pollutants can induce IL-6 secretion of L02 cells in a dose-and time-dependent manner, induce inflammatory reaction, induce apoptosis and lead to hepatocyte damage.
