二十八烷醇的制备及其对机体能量代谢的影响和机理研究

【摘要】:本文详细论述了以蜂蜡为原料制备二十八烷醇的工艺,并进行优化;其次以SD大鼠为实验模型,研究了二十八烷醇对于SD大鼠生长性能、血液生化指标、激素分泌量等的影响;最后通过分子生物学实验,研究了二十八烷醇对于SD大鼠机体参与能量代谢过程基因的表达量的影响,深入研究了对于大鼠能量代谢的调控机制。 试验一:实验首先将精制蜂蜡用醇相皂化法处理,研究了皂化过程中所需要的皂化温度、皂化时间、NaOH乙醇溶液浓度、固液比四个单一因素对于皂化提取过程的影响,确定了在单因素条件下的最佳条件,然后通过正交试验设计确定最佳的皂化条件为皂化温度95℃、皂化时间3.5h、NaOH乙醇溶液浓度为8%、固液比为1:8。然后经过分子蒸馏技术纯化可以制得最高纯度89.78%的二十八烷醇。 试验二:以SD大鼠为模型,,在饲料中添加了不同浓度的二十八烷醇,研究了二十八烷醇对于SD大鼠生长性能、生理生化参数、激素分泌量的影响。实验选用了48只健康的SD大鼠(雌雄各半),按照体重一致原则分为4组:实验1组(生理盐水空白对照组)、实验2组(低二十八烷醇浓度组)、实验3组(中二十八烷醇浓度组)、实验4组(高二十八烷醇浓度组)。大鼠在饲养期间自由采食,饮用蒸馏水,试验时间为30天。记录采食量、体重等各项指标。实验后期采集大鼠血液、肝脏、肌肉等组织样品,荧光定量GLUT-4、StAR、GS、AMPK等多个基因的表达。试验结果显示: 1相对于生理盐水空白对照组,饲喂高、中、低三个不同浓度的二十八烷醇乳化剂后显著提高SD大鼠的体重、日增重和饲料转化率,对于生理生化参数影响显著;SD大鼠生长激素、胰高血糖素等分泌量均明显增加。 2相对于生理盐水空白对照组,饲喂高、中、低三个不同浓度的二十八烷醇乳化剂后SD大鼠体内总胆固醇、低密度脂蛋白显著降低,高密度脂蛋白明显增加;中浓度实验组的调节作用最佳。 3相对于生理盐水空白对照组,饲喂高、中、低不同浓度的二十八烷醇乳化剂后SD大鼠肝脏和肌肉组织中GLUT-4、STAR、GS、AMPK等多个基因均高于对照组,且中浓度实验组表达量增加最明显。
[Abstract]:In this paper, the preparation process of 28 alkanol from beeswax was discussed in detail and optimized. Secondly, the effects of 28 alkanol on growth performance, blood biochemical index and hormone secretion of SD rats were studied using SD rats as experimental model. Finally, the effects of 28 alkanol on the expression of genes involved in energy metabolism in SD rats were studied by molecular biological experiments, and the regulation mechanism of energy metabolism in rats was also studied. Experiment 1: the refined beeswax was first treated by alcohol phase saponification method. The effects of four single factors, such as saponification temperature, saponification time, concentration of NaOH ethanol solution and solid-liquid ratio, on the saponification extraction process were studied. The optimum conditions were determined under the single factor condition, and the optimum saponification conditions were determined by orthogonal design: saponification temperature 95 鈩
