X射线对人周围血淋巴细胞Gadd45 mRNA表达影响
[Abstract]:Objective to analyze the effects of different doses and time of irradiation on the growth inhibition of human peripheral blood lymphocytes and the relative expression of DNA damage gene 45 (Gadd45) mRNA, and to explore the possibility of Gadd45 gene as a biological dosimeter. Methods 1 the dose-effect relationship was studied. The peripheral blood samples of healthy people were irradiated in vitro with the doses of 0.000.10 ~ 0.25 ~ 0.50 ~ 1.00 ~ 1.00 ~ 2.00 ~ 3.00 and 5.00 Gy respectively, and the relationship between time and effect was studied. Peripheral blood samples of healthy people were irradiated with 2. 00 Gy dose of X ray in vitro. Lymphocytes were isolated and cultured for 24 h and 48 h, respectively. The relative expression of Gadd45 mRNA was detected by real-time fluorescence quantitative polymerase chain reaction. Results within the dose range of 0.50 Gy, the relative expression of Gadd45 mRNA increased with the increase of irradiation dose. The regression equation was 1.056 9 7.132 2 x [determination coefficient (R2) = 0.992 2]. P0.01. The relative expression of Gadd45 mRNA in 0.50 Gy group reached the peak value, 4.53 times of that in 0.00 Gy group. Within the dose range of 0.50 Gy 5.00 Gy, the relative expression level of Gadd45 mRNA decreased, and the relative expression level of Gadd45 mRNA increased with the increase of irradiation time at 3h 12 h after 22.00 Gy dose irradiation. The regression equation was Yi-6.366 0 4.965 x (R _ 2N _ (0.932) P _ (0.05). The relative expression level of Gadd45 mRNA reached the peak at 12 h after irradiation, which was 57.68 times of that in 0 h group, and the relative expression level of Gadd45 mRNA decreased significantly at 24 h after irradiation 48 h after irradiation, and 2.08 times as much as 0 h at 48 h after irradiation. Conclusion the expression of Gadd45 mRNA can be upregulated by X-ray irradiation, which has a good dose-effect and time-effect relationship within a certain dose and time range, but the Gadd45 gene as a new biological dosimeter needs further study and verification.
