柚子籽油营养分析及其微胶囊制备

【摘要】:柚子籽油是芸香科柑橘亚科柑橘属植物柚子(Citrus paradise Macf.)的种子油脂,尚未得到有效利用。本研究对柚子籽油进行了营养分析,研究表明它富含不饱和脂肪酸以及柠檬苦素等物质,具有较高的营养价值。由于柚子籽油本身具有不良苦味,而且所含营养物质不稳定,易氧化,为了掩盖柚子籽油的苦味并提高其稳定性,本研究通过饱和水溶液法制备β-CD/柚子籽油微胶囊,以期为柚子籽油的合理开发利用提供一定参考。主要研究内容及结果如下: 1.评价柚子籽油营养分析 柚子籽油的相对密度0.968g/mL,折光指数1.471,过氧化值5.63mmol/kg,酸值1.06mgKOH/g,皂化值196.46mg KOH/g,碘值97.896g/100g,各理化指标都比较适宜。GC分析表明柚子籽油中含有66.95%的不饱和脂肪酸,其中LA35.4%(其中CLA0.58%)、油酸26.38%、α-亚麻酸3.92%;含有33.02%的饱和脂肪酸,其中棕榈酸29.60%、硬脂酸2.86%。 FT-IR分析表明,柚子籽油主要有10个吸收峰,表现在:3750-3000cm-1区间的O-H伸缩振动,3000-2800cm-1区间的饱和C-H伸缩振动,1900-1630cm-1区间的羰基伸缩振动,1475-1300cm-1区间的饱和C-H面内弯曲振动,1000-650cm-1区间的不饱和C-H面外弯曲振动。由分析结果可推断柚子籽油含有不饱和脂肪酸,也可进一步为柚子籽油鉴别、营养分析提供信息。 2.HPLC法检测柚子籽油中柠檬苦素含量 建立了柚子籽油中柠檬苦素的HPLC检测方法,色谱条件:色谱柱为Agilent Zorbax Extend-C18(4.6mm×250mm,5μm),流动相为乙腈-水(45:55),流速0.8mL/min,检测波长215nm,柱温30℃。结果表明柠檬苦素在48~384μg/mL (R2=0.9997)浓度范围内线性关系良好,加样平均回收率为99.87%, RSD=1.33%(n=3)。在此条件下测得柚子籽油中柠檬苦素含量为327.61μg/g 3.β-CD/柚子籽油微胶囊的制备 以饱和水溶液法制备p-CD/柚子籽油微胶囊。研究表明投料比(p-CD/柚子籽油,g/mL)、包埋时间、包埋温度对微胶囊包埋率影响程度大小为:包埋温度包埋时间投料比。在单因素试验基础上通过响应面分析法优化制备工艺,确定最佳工艺为:投料比4.5,包埋时间132min,包埋温度47℃,在此工艺条件下包埋率达92.18%。4.p-CD/柚子籽油微胶囊质量评价 物相鉴定试验表明β-CD已经成功包埋柚子籽油形成微胶囊,且包埋过程中柚子籽油化学成分未发生变化,扫描电镜下可见微胶囊颗粒较圆整,表面附着有小颗粒。微胶囊平均粒径为1.58pm、含水率3.19%、密度0.452g/cm3、溶解度70.31%、休止角36°;微胶囊载油量为74.32%;热稳定性试验表明p-CD/柚子籽油微胶囊质量减少量低于柚子籽油;预测货架期可达到182d,高于柚子籽油(74d),因此,可以判断柚子籽油经包埋后对热稳定性提高。 综上所述,柚子籽油具有较高营养价值,且饱和水溶液法操作简便,所需设备要求不高,成本较低,制得的β-CD/柚子籽油微胶囊包埋率较高,热稳定性良好,具有一定推广开发价值。

It can be concluded that pomelo seed oil contains unsaturated fatty acids, and it can also provide information for identification and nutrition analysis of pomelo seed oil.
