DHA干预对GDM小鼠胎盘脂肪酸转运蛋白基因表达的影响
[Abstract]:Objective to investigate the effect of 22 carbohexaenoic acid (DHA) intervention on the expression of fatty acid transporter gene in placenta of (GDM) mice with gestational diabetes mellitus (GDM). Methods 36 mice were selected, 12 of which were used as blank control (CN), 24 mice were injected intraperitoneally with streptozotocin to establish GDM model, GDM mice were randomly divided into two groups: (GDM DHA) supplemented with DHA and (GDM). As control group. From the 10th day to the 18th day, the rats in the GDM DHA group were given DHA 500 mg/kg, and the other two groups were given the same amount of solvent. The blood glucose and body weight of the three groups were recorded. The serum and placenta of the pregnant rats were collected on the 18th day. The activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-px) in maternal serum were measured by hydroxylamine method and colorimetric method, and the expression level of fatty acid transporter gene in placenta was measured by RT-PCR method. Results (1) the results of ANOVA of repeated measurements showed that the weight of pregnant rats and the difference of blood glucose in different time periods were statistically significant (P0.05). There was significant difference in blood glucose between different groups of pregnant rats (P0.05). (2). There was significant difference in SOD activity and GSH-px activity level among the three groups (P0.05). Compared with CN group, the activity level of SOD and GSH-px in GDM group was significantly lower than that in CN group (P0.05). (3), compared with CN group (P0.05). (3), after intervention with), DHA, the activity level of SOD in mice was significantly higher than that in CN group. GDM down-regulated the expression of peroxisome proliferator-activated receptor 纬 (PPAR 纬) mRNA and up-regulated the expression of fatty acid binding protein (FABP-pm) and fatty acid transporter-6 (FATP-6 mRNA). After DHA intervention, the expression of FABP-pm,FATP-6 mRNA was down-regulated. Conclusion DHA can increase the activity of SOD and down-regulate the expression of placental FABPpm,FATP-6 mRNA in GDM mice.
