有氧运动对MI后大鼠VEGF、bFGF分泌及心脏血管新生的影响研究

【摘要】:研究目的:探讨不同时长有氧运动对心肌梗死(MI)后大鼠血管内皮生长因子(VEGF)、碱性成纤维细胞生长因子(bFGF)分泌情况和碱性成纤维细胞生长因子受体(FGFR)的mRNA表达水平,以及对心脏血管新生的影响。研究方法:选用雄性8周龄SD大鼠40只,随机分为假手术组(Sham)、心肌梗死组(MI)、梗死+运动2周组(ME2),梗死+运动8周组(ME8),每组10只。以冠状动脉左前降支(LAD)结扎术构建MI模型,ME组术后1周开始运动。运动方案为:适应性运动1周后,每天10 m/min热身10 mmin,然后以高强度20 m/min运动8min,再以低强度15 m/min运动2 min,高低强度交替4个循环,最后20 m/min运动8min,10 m/min运动2min后结束。每天训练1次,5 d/w,共训练8w。以二维超声心动图检测大鼠心脏室壁厚度、射血功能;大鼠心肌组织血管新生情况分别采用透射电镜扫描、Westernblot检测心肌组织CD34的表达水平来衡量;采用ELISA的方法检测大鼠血浆、心肌组织VEGF、 bFGF分泌水平,RT-PCR检测FGF-R的mRNA水平。心室研究结果:(1)心梗使大鼠的心脏重量损失,有氧运动后心脏重量损失有明显的改善作用,有氧运动8周使心重/体重比值(HW/BW)明显增加(P≤0.05)、左心室重量/体重比值明显增加(P≤0.05)。(2)心肌梗死手术使大鼠心脏心室壁变薄,射血分数(EF)显著降低(P≤0.01);有氧运动使MI大鼠的室壁厚度增加、射血功能加强,其中ME8组舒张期左室后壁厚度(LVPWd)与舒张期室间隔厚度之和明显高于MI组(P0.05), ME8组EF比MI组明显升高(P≤0.05)。(3)电镜观察结果显示,MI使心肌组织血管内皮细胞损伤、间隙增大,基底膜不完整,ME2和ME8组均能见到新生的微小血管。CD34的Westernblot结果显示,ME2组的CD34表达水平比MI组明显升高(P≤0.05),提示血管新生程度活跃。(4)有氧运动使血浆和心肌组织的VEGF分泌水平明显升高,其中ME2组血浆的VEGF水平比MI组明显升高(P≤0.05),ME8组血浆的VEGF水平比MI组明显著升高(P≤0.01),且明显高于ME2组(P≤0.05);ME2组(P≤0.01)和ME8组(P≤0.01)心肌组织的VEGF水平都显著高MI组。(5)有氧运动使血浆和心肌组织的bFGF分泌水平明显升高,其中ME2组(P≤0.01)和ME8组(P≤0.01)血浆的bFGF水平比MI组都有显著升高;ME2组(P≤0.01)和ME8组(P≤0.01)心肌组织的bFGF水平也都显著高于MI组。(6)有氧运动使bFGF-R的mRNA表达水平明显升高,ME2组(P≤0.05)、ME8组(P≤0.05)与MI组相比,bFGF-R的mRNA水平都有明显升高。研究结论:(1)有氧运动有利于MI大鼠心脏舒缩功能的改善;(2)有氧运动能促进MI大鼠心肌组织血管新生;(3)有氧运动可能是通过上调VEGF、 bFGF的分泌水平,以及bFGF-R的mRNA表达水平来促进血管新生的。
[Abstract]:Objective: to investigate the effects of aerobic exercise on vascular endothelial growth factor (VEGF) after myocardial infarction (MI) in rats. The secretion of basic fibroblast growth factor (bFGF) and the mRNA expression of basic fibroblast growth factor receptor (BFGFR). Methods: forty male 8-week-old SD rats were randomly divided into sham-operated group and myocardial infarction group (MIM). The MI model was established by ligation of left anterior descending branch of coronary artery (LAD). The ME group began to exercise 1 week after the operation. The exercise plan was: after 1 week of adaptive exercise, 10 m / min, 10 min / min daily, and then 8 minutes of high intensity 20 m / min exercise. Low intensity 15m / min exercise for 2 min, high and low intensity alternating 4 cycles, and last 20 m / min exercise 8 minutes. 10 m / min exercise lasted 2 minutes. The rats were trained once a day for 5 d / w for 8 weeks. The ventricular wall thickness and ejection function were measured by two dimensional echocardiography. The expression of CD34 in myocardial tissue was measured by transmission electron microscope scanning and Western blot. The levels of VEGF and bFGF in plasma and myocardial tissue of rats were measured by ELISA. RT-PCR was used to measure the mRNA level of FGF-R. Ventricular study results showed that myocardial infarction could significantly improve cardiac weight loss after aerobic exercise in rats.
