单次大剂量电子线照射SD大鼠胃的损伤观察

【摘要】:目的:使用单次大剂量电子线照射SD大鼠胃,模拟胃部术中放疗(Intraoperative radiotherapy,IORT),观察大鼠胃照射后的损伤变化及修复情况,为大剂量术中放疗提供动物实验理论性依据。方法:38只八周龄健康雄性SD大鼠随机分为两组:正常对照组(A组)8只,实验组(B组)30只。使用专利发明“小动物放射线精确照射试验台[1]”,配合电子线托架,自制1cm厚度,3×3cm2照射野铅块,6MeV电子线单次20Gy准确照射B组大鼠胃。动态观察大鼠照射后的一般情况、饮食量、饮水量、体重的变化和胃组织粘膜病理学改变。所得体重和饮食量数据以均数±标准差,分别用SPSS 17.0软件处理,分析结果。结果:B组大鼠经20Gy电子线单次照射后,受照区体毛脱落稀疏,前两周活动减少,精神萎靡,死亡率10%;饮食量在照射后八天内较A组减少(P0.05);体重在第一周内明显下降,第二周体重开始增加,体重增加最快在第四周至第六周。第八周时大鼠饮食量已和对照组没有差异(P0.05),两组体重比较在第六周时没有差异(P0.05)。A组饮食量和体重一直相对平稳增长。病理大体观B组大鼠胃,未见穿孔、坏死,胃与周围组织均有纤维性粘连,在照射后第14天时粘连最重且颜色鲜红,第56天时最轻且色灰黄。HE染色检查:B组照射后第14天见,胃粘膜糜烂、粘膜下高度水肿。照射后28天见,粘膜下水肿,纤维化、腺体萎缩。第56天见,胃腺体萎缩,炎性纤维素性渗出。A组胃组织结构完整,粘膜颜色鲜红、细胞排列整齐、无糜烂,腺体无萎缩,无纤维化及炎细胞浸润,粘膜下层、肌层和浆膜层均正常。结论:SD大鼠胃可耐受6MeV电子线单次20Gy照射,无穿孔、坏死等严重并发症发生。在照射后,两周内急性反应最重,对体重及饮食影响大,此时,可能是放射性粘膜损伤保护性治疗的较佳时机。第八周时已完全形成纤维性修复,提示在照射能量和剂量还可进一步加大,探索大鼠胃受照射能承受的更高剂量。
[Abstract]:Objective: to observe the changes and repair of intragastric radiation induced by intragastric irradiation in SD rats. Methods Thirty eight healthy male Sprague-Dawley rats were randomly divided into two groups: normal control group (n = 8), control group A (n = 8) and control group A (n = 8). Experimental group B (n = 30). Using patent invention "small animal radiation precise irradiation test stand [1]", with electronic wire carrier, The stomach of rats in group B was accurately irradiated with 1 cm thick and 3 脳 3 cm ~ 2 electron rays of 6 MeV electron rays at a single dose of 20 Gy. The general situation, diet and drinking quantity of rats after irradiation were observed dynamically. Weight changes and pathological changes of gastric mucosa. The weight and diet data were treated with SPSS 17.0 software in the mean 卤standard deviation. Results the body hair in the irradiated area of rats in group B was sparse after 20 Gy electron ray irradiation. During the first two weeks, activity decreased, mental retardation, mortality rate 10. 5% lower than group A in eight days after irradiation, weight decreased significantly in the first week, and weight began to increase in the second week. The maximum weight gain was from the fourth week to 6th weeks. At 8th weeks, there was no difference between the rats and the control group (P 0.05), but there was no difference between the two groups at 6th weeks. The diet and body weight of group A were relatively stable. There was no perforation, necrosis, fibrous adhesion between stomach and surrounding tissue. On the 14th day after irradiation, the adhesion was the heaviest and the color was bright red. On the 56th day, the grayish yellow color was the lightest and grayish yellow. He staining showed that the gastric mucosa was erosive on the 14th day after irradiation in group B. Submucosal high edema. 28 days after irradiation, submucosal edema, fibrosis, atrophy of glands. 56 days, gastric gland atrophy, inflammatory cellulose exudation. Group A gastric tissue structure integrity, mucosal color bright red, cells arranged neatly, no erosion, No atrophy, no fibrosis, no inflammatory cell infiltration, normal submucosal, muscular and serous layers were found in the stomach of the rats. Conclusion the stomach of the SD rats can tolerate a single dose of 20 Gy irradiation with 6 MeV electron lines, no perforation, necrosis and other serious complications. The acute reaction was the most serious in two weeks, which had a great influence on body weight and diet. At this time, it may be a better time for protective treatment of radiation mucosal injury. At 8th weeks, the fibrous repair has been completely formed, suggesting that the radiation energy and dose can be further increased. To explore the higher dose of rat stomach irradiation.
