姜黄素对力竭运动大鼠肾损伤的保护作用及其机制的研究

【摘要】:目的:探讨姜黄素对力竭运动大鼠肾脏损伤的保护作用及其机制。 方法:SPF级健康雄性SD大鼠56只,体重210±10g,按随机数字法分为正常对照组(C)、力竭模型组(E)、力竭+氯沙坦组(阳性对照组)(LE)、力竭+低剂量姜黄素组(CMN1)、力竭+中剂量姜黄素组(CMN2)、力竭+高剂量组姜黄素(CMN3)、姜黄素对照组(CMN),每组8只。力竭+姜黄素组每天灌胃不同剂量姜黄素溶液(低剂量100mg/kg、中剂量200mg/kg、高剂量300mg/kg);阳性对照组每天氯沙坦灌胃20mg/kg;姜黄素对照组灌胃剂量为200mg/kg/d;正常对照组常规条件下饲养,正常饮食,不做任何运动;各运动组进行力竭运动训练,每周记录各组大鼠体重,4周后,各组大鼠取血清、血浆、肾脏组织低温保存,HE染色光镜下观察肾脏形态结构变化,分光光度法检测血清和肾脏组织BUN、Cr、SOD及MDA的含量,采用双抗体一步夹心法酶联免疫吸附(ELISA)检测血清及肾组织中TNF-α、 IL-1β、IL-18、TGF-β的含量,比色法检测caspase-3的活性。 结果: (1)姜黄素可明显延长力竭运动大鼠力竭运动持续时间,与力竭模型组相比差异具有统计学意义(p0.05) (2)与力竭模型组相比,应用姜黄素后,血清及肾组织中MDA、BUN、Cr含量明显降低(p0.05), SOD(?)舌性显著升高(p0.05);三种不同剂量姜黄素组比较,中剂量组(200mg/kg)对BUN、Cr、SOD及MDA影响效果优于低、高剂量组(p0.05);阳性对照组与中剂量姜黄素组比较,差异无统计学意义(p0.05)。 (3)姜黄素干预后,大鼠肾组织及血液中TNF-α、IL-1β、IL-18、TGF-β及caspase3水平较力竭模型组显著降低(p0.05);中剂量组(200mg/kg)与低、高剂量组相比均有差异,对TNF-α、IL-1β、IL-18、 TGF-β及caspase3影响效果显著优于二组(p0.05);阳性对照组与中剂量组比较,差异无统计学意义(p0.05)。 (4)光学显微镜下观察肾组织切片,姜黄素干预的力竭运动大鼠肾脏肾损伤程度均比力竭模型组减轻。 结论: (1)姜黄素可显著延长力竭持续时间。 (2)姜黄素可能通过提高抗氧化酶活性,清除自由基等作用缓解大鼠力竭运动造成的肾脏损伤。 (3)姜黄素可能通过抑制促炎症相关因子TNF-α、IL-1β、IL-18的生成,拮抗促纤维生长因子TGF-β的表达,抑制caspase-3活性,对力竭运动大鼠肾损伤起保护作用。 (4)中剂量的姜黄素(200mg/kg)较其他两种剂量有更明显的肾脏保护作用。
[Abstract]:Objective: to investigate the protective effect and mechanism of curcumin on kidney injury in exhaustive exercise rats. Methods 56 healthy male Sprague-Dawley rats of SPF grade, The body weight was 210 卤10 g. The rats were randomly divided into three groups: normal control group (Con), exhausted model group (EJ), exhausted losartan group (positive control group), exhausted curcumin group (low dose curcumin group), exhausted middle dose curcumin group (CMN2), exhausted high dose group (curcumin CMN3). Different doses of curcumin solution (100mg / kg, middle dose 200mg / kg, high dose 300mg / kg) were given daily in the exhausted curcumin group, 20 mg / kg daily in the positive control group, 20 mg / kg in the curcumin control group, and 200 mg / kg / d in the curcumin control group. The normal control group was fed under normal conditions. Every exercise group had exhaustive exercise training. After 4 weeks of exercise, the rats in each group were collected serum, plasma and kidney tissue to observe the changes of renal morphology under light microscope with HE staining. The contents of BUNC and MDA in serum and renal tissue were detected by spectrophotometry. The levels of TNF- 伪-尾 IL-1 尾 IL-18 TGF- 尾 and the activity of caspase-3 in serum and renal tissue were detected by double antibody one step sandwich enzyme-linked immunosorbent assay (Elisa). Results:. 1) curcumin significantly prolonged the duration of exhaustive exercise in rats, and the difference was statistically significant compared with the exhaustive model group (P 0.05). (2) compared with the exhaustive model group, the content of MDA-BUNCr in serum and renal tissue decreased significantly after curcumin administration. Compared with three different doses of curcumin, the effect of middle dose group (200 mg 路kg ~ (-1)) on MDA and SOD was better than that of high dose group (P 0.05), but there was no significant difference between positive control group and middle dose group (P 0.05). (3) after curcumin intervention, the levels of TNF- 伪 IL-1 尾 IL-18TGF- 尾 and caspase3 in renal tissue and blood of rats were significantly lower than those of exhausted model group, and the levels of TNF- 伪 IL-1 尾 IL-18TGF- 尾 and caspase3 in middle dose group were significantly lower than those in low dose group and high dose group. The effect on TNF- 伪 IL-1 尾 IL-18, TGF- 尾 and caspase3 was significantly better than that in the two groups (P 0.05), but there was no significant difference between the positive control group and the middle dose group (P 0.05). (4) the renal tissue sections were observed under optical microscope. The degree of kidney injury in rats treated with curcumin was less than that in exhausted model group. Conclusion:. Curcumin significantly prolonged the exhaustion duration. 2) curcumin may alleviate the kidney injury induced by exhaustive exercise by increasing the activity of antioxidant enzymes and scavenging free radicals. 3) curcumin may inhibit the expression of TGF- 尾, inhibit the expression of TGF- 尾 and inhibit the activity of caspase-3 by inhibiting the production of TNF- 伪 and IL-1 尾 and IL-18. Curcumin may play a protective role on renal injury in rats after exhaustive exercise. The middle dose of curcumin 200 mg / kg had a more obvious renal protective effect than the other two doses.
