基于~1H-NMR的SD大鼠弥漫性轴索损伤的代谢组学研究

【摘要】:目的探讨基于1H-NMR的检测技术应用于SD大鼠DAI血浆代谢组学研究的可行性,并寻找筛选早期诊断DAI的可能的特异性生物标志物。 方法采用改良的Marmarou A方法建立SD大鼠DAI模型,将所有实验动物随机分为5组(空白对照组(n=6)、手术1天对照组(n=6)、打击致伤1天组(n=8)、手术3天对照组(n=6)、打击致伤3天组(n=8)),收集各组SD大鼠右心室血液,制备成血浆后通过1H-NMR检测,采用PCA、PLS-DA和OPLS-DA进行数据处理分析。 结果与对照组相比,DAI组SD大鼠血浆中1H-NMR有明显差异,各组血浆样本间有32种分子代谢物有显著差异,DAI1天时Glu明显升高,3天时Lac显著升高,致伤到伤后3天脂质成分(VLDL和LDL)一直显著降低。 结论1H-NMR技术是研究DAI代谢组学的有效技术手段。分析鉴定出的32种代谢物及其变化规律或可为DAI早期诊断特异性生物标志物的筛选提供可靠的实验依据。
[Abstract]:Objective to explore the feasibility of using 1H-NMR based detection technique in the study of plasma metabolism in SD rats with DAI, and to find out the possible specific biomarkers for early diagnosis of DAI. Methods the DAI model of SD rats was established by modified Marmarou A method. All experimental animals were randomly divided into five groups: control group (control group), control group (control group), control group (control group), control group (control group), control group (control group), control group (control group), control group (control group), control group (control group), control group (control group), control group (n = 6) and control group (n = 8), respectively. Blood samples from right ventricle of SD rats in each group were collected, and plasma was prepared for detection by 1H-NMR. Data processing and analysis were carried out by PCAA PLS-DA and OPLS-DA. Results compared with the control group, there were significant differences in plasma 1H-NMR in SD rats, and there were significant differences in 32 kinds of molecular metabolites among plasma samples of each group. At day 1, Glu was significantly increased and Lac was significantly increased at day 3. VLDL and LDL decreased significantly 3 days after injury. Conclusion the 1H-NMR technique is an effective technique for the study of DAI metabolomics. The analysis and identification of 32 metabolites and their changes may provide a reliable experimental basis for the screening of specific biomarkers for early diagnosis of DAI.
