耐力运动及环孢霉素A对大鼠骨骼肌纤维组成的影响

【摘要】:研究目的:通过耐力运动及注射环孢霉素A(CsA)干预钙调神经磷酸酶(CaN)的信号通路,探讨对大鼠骨骼肌纤维组成的影响。 研究方法:以雄性SD大鼠为研究对象,分组进行适应性训练,将SD大鼠随机分为四组:对照组(C组)、CsA组(S组)、运动CsA组(ST组)、单纯运动组(T组),每组10只。四组SD大鼠在第1~2周进行两周的适应性训练,第3~第15周进行耐力训练, S组、ST组在第13至15周进行皮下注射CsA15mg/kg/day(共3周)。在停止训练后的第二天取SD大鼠的比目鱼肌和趾长伸肌,用SDS-PAGE电泳测肌球蛋白重链(MHC)组成。用南京建成的琥珀酸脱氢酶试剂盒测试SDH的活性和用定磷法测CaN的活性,并用Western blotting法测活化T细胞核因子(NFATc1)的蛋白含量。 研究结果: SDH: ST组与C组相比有非常显着性升高(P<0.05);ST组与C组相比有非常显着性升高(P<0.01)。 肌球蛋白浓度:大鼠比目鱼肌(SOL)肌球蛋白浓度比较, S组与C组相比有显着性下降(P<0.05);ST组与C组相比显着性下降(P<0.05)。大鼠趾长伸(EDL)各组肌球蛋白的浓度无明显差异。 CaN:大鼠比目鱼肌(SOL)的CaN活性,S组与C组相比有显著性下降(P<0.05);ST组与C组相比有非常显著性下降(P<0.01);ST组与T组相比有显著性下降(P<0.05)。大鼠趾长伸肌(EDL)的CaN的活性, T组与C组相比有非常显著性升高(P<0.01);ST组与S组相比有显著性下降(P<0.05);ST组与T组相比有非常显著性下降(P<0.01)。 MHC:大鼠比目鱼肌MHCⅠ,ST组与C组相比具有显著性升高,MHCⅡa显著性下降(P<0.05)。大鼠趾长伸肌MHCⅡx, S组与C组相比有非常显著性升高,MHCⅡd非常显著性下降(P<0.01)。大鼠趾长伸肌MHCⅡx, ST组与S组相比具有非常显著性下降MHCⅡd非常显著性升高(P<0.01)。 NFATC1:大鼠比目鱼肌NFATC1, T组与C组相比有显著性下降(P<0.05);ST组与C组相比具有非常显著性下降(P<0.01);ST组与S组相比有非常显著性下降,,(P<0.01)。 研究结论: 1.注射CsA可导致大鼠饮食减少、体重下降,并伴有皮下组织囊肿和溃烂。 2.耐力运动增强了有氧氧化酶的活性,使大鼠骨骼肌SDH得到进一步的发展。CsA的干预并没有影响SDH的活性变化。 3.耐力运动和注射CsA均可以引起大鼠慢肌比目鱼肌的肌球蛋白降解。 4.注射CsA可以抑制CaN的活性,耐力运动可以引起快肌趾长伸肌的CaN活性提高,但对对慢肌比目鱼肌的CaN活性没有影响,说明不同肌纤维注射CsA干预CaN的程度不同。 5.耐力运动及注射CsA可以使慢肌比目鱼肌的MHCⅠ增加、MHCⅡa减少,快肌趾长伸肌MHCⅡx减少、MHCⅡd增加。 6.耐力运动使慢肌比目鱼肌中Ca2+浓度提高,引起CaN的活性提高,使大量NFATC1去磷化进入胞核内。注射CsA抑制了CaN的表达使NFATC1没有进入细胞核中。耐力运动及CsA共同作用使比目鱼肌使胞浆中NFATC1大量入胞核。
[Abstract]:Objective: to investigate the effects of endurance exercise and cyclosporine (CsA) injection on the signal pathway of calmodulin phosphatase (Ca) in skeletal muscle of rats. Methods: male Sprague-Dawley rats were divided into groups for adaptive training. Sprague-Dawley rats were randomly divided into four groups: control group C, CSA group S group, exercise CsA group St group, exercise group T group with 10 rats in each group. Endurance training was conducted from week 3 to week 15, and group S was given subcutaneous injection of CSA 15 mg / kg / day at the 13th to 15th weeks. The soleus muscle and extensor digitorum longus muscle of SD rats were taken on the second day after stopping training. The composition of myosin heavy chain (MHC) was measured by SDS-PAGE electrophoresis. The activity of SDH was tested by the succinate dehydrogenase kit built in Nanjing, the activity of CaN was measured by the method of fixed phosphorus, and the protein content of activated T cell nuclear factor (NFATc1) was measured by Western blotting method. Results of the study:. SDH: there was a significant increase in SDH between St group and C group (P < 0.05). Myosin concentration: compared with group C, the concentration of myosin in group S was significantly lower than that in group C (P < 0.05) and that in group St was significantly lower than that in group C (P < 0.05). There was no significant difference in the concentration of myosin between group S and group C (P < 0.05), but there was no significant difference in the concentration of myosin between group S and group C (P < 0.05). The activity of CaN in group S was significantly lower than that in group C (P < 0.05). The activity of CaN in group St was significantly lower than that in group C (P < 0.01). The activity of CaN in extensor digitorum longus of rats was significantly decreased compared with group T, and the activity of CaN in group S was significantly lower than that in group C (P < 0.01). The activity of CaN in extensor digitorum longus in S group was significantly lower than that in group C (P < 0.05), and that in group T and T was significantly lower than that in group C (P < 0.01). There was a very significant increase in group C (P < 0.01) and a significant decrease in group S (P < 0.05) compared with group T (P < 0.01). MHC: compared with C group, MHC 鈪
