跑台运动对去卵巢应激大鼠行为学改变及前额叶皮质单胺类神经递质水平和BDNF表达的影响

【摘要】:目的观察4周跑台运动对慢性应激致去卵巢大鼠抑郁、焦虑行为及空间学习记忆能力和前额叶皮质(Prefrontal Cortex,PFC)单胺类神经递质水平及脑源性神经营养因子(Brain-Derived Neurotrophic Factor,BDNF)表达的影响。方法 40只SD大鼠随机分为假手术组、去卵巢组、去卵巢应激组及去卵巢应激运动组,除了假手术组大鼠,其余3组大鼠采用卵巢摘除建立大鼠围绝经期模型,再通过慢性不可预知性应激(chronic unpredictable stress,CUMS)建立抑郁模型,同时,应激运动组大鼠进行4周跑台运动。运动及应激结束后通过旷场实验、高架迷宫实验、明暗箱探索实验及水迷宫实验等方法测试大鼠行为学指标变化,采用高效液相-电化学检测法检测前额叶皮质去甲肾上腺素(norepinephrine,NE)、多巴胺(dopamine,DA)及5-羟色胺(5-hydroxy trptamine,5-HT)的含量,用免疫组织化学结合图像半定量方法对前额叶皮质BDNF神经元的数量、面积进行测量和分析。结果卵巢摘除及慢性不可预知应激增强了大鼠的抑郁行为及焦虑行为,学习记忆能力下降,行为学表现为旷场试验中大鼠穿越格数及直立次数均显著减少,高架迷宫实验中OE%、OT%及(开放臂+闭合臂)均显著下降,明暗箱实验中大鼠明箱时间缩短,水迷宫实验中学习记忆能力下降;血清皮质醇(cortisol,Cort)水平增加,前额叶皮质单胺类神经递质DA、NE、5-HT含量均显著下降,BDNF表达减弱。而长期跑台运动可以显著改善去卵巢抑郁模型大鼠的抑郁样行为及增强学习记忆能力,血清Cort水平下降,前额叶皮质单胺类神经递质DA、NE、5-HT含量增加,BDNF表达增强。结论跑台运动可改善去卵巢抑郁模型大鼠的抑郁及焦虑行为,增强大鼠学习记忆能力,机理可能与长期跑台运动降低去卵巢抑郁大鼠Cort水平、拮抗HPA轴功能亢进、上调前额叶皮质BDNF的表达、有效调整中枢神经递质紊乱有关。
[Abstract]:Objective to observe the effects of treadmill exercise on depression, anxiety behavior, spatial learning and memory ability, monoamine neurotransmitter level in Prefrontal Cortexa cortex and the expression of Brain-Derived Neurotrophic factorial neurotrophic factor (BDNF) in ovariectomized rats induced by chronic stress. Methods Forty SD rats were randomly divided into three groups: ovariectomized stress group, ovariectomized stress group and ovariectomized exercise group. The depression model was established by chronic unpredictable stress (chronic unpredictable stress CUMS). At the same time, the rats in the stress exercise group underwent treadmill exercise for 4 weeks. After exercise and stress, the behavioral indexes of rats were measured by open field experiment, elevated maze test, open and dark box exploration experiment and water maze test. The contents of norepinephrine (NE), dopamine (DA) and 5-hydroxy trptamine 5-HT (5-HT) in prefrontal cortex were detected by high performance liquid chromatography (HPLC) and electrochemical method. The number of BDNF neurons in prefrontal cortex was determined by immunohistochemistry and image semi-quantitative method. The area is measured and analyzed. Results Ovariectomized and chronic unpredictable stress enhanced the depression behavior and anxiety behavior, decreased learning and memory ability, and significantly decreased the number of traversals and the number of upright rats in open field test. In the elevated labyrinth experiment, the OEO% and open arm closed arm decreased significantly, the open box time shortened, the learning and memory ability decreased and the serum cortisol level increased. The expression of BDNF decreased significantly in prefrontal cortex monoamine neurotransmitters. Long-term treadmill exercise could significantly improve the depressive behavior and enhance the learning and memory ability of ovariectomized depression rats. The serum Cort level decreased and the prefrontal cortex monoamine neurotransmitter DANE-5-HT increased. Conclusion treadmill exercise can improve the depression and anxiety behavior of ovariectomized depression rats and enhance the learning and memory ability of the rats. The mechanism may be associated with long-term treadmill exercise to reduce the level of Cort and antagonize the hyperfunction of HPA axis in ovariectomized depression rats. The up-regulation of BDNF expression in prefrontal cortex is related to the regulation of central neurotransmitter disorder.
