放射线照射大鼠涎腺组织后对Rad50蛋白表达及涎腺组织凋亡的影响

【摘要】:目的:研究不同剂量C060γ射线照射大鼠涎腺组织后Rad50蛋白的表达变化情况及其对涎腺组织凋亡的影响。 方法:选取近交系雄性Wistar大鼠60只,按照随机数字表法分成6组:对照组(未进行放射,即0Gy组),放射组3Gy组、6Gy组、9Gy组、12Gy组、15Gy组,每组10只。每次3Gy,隔天一次,累积剂量分别为3Gy组、6Gy组、9Gy组、12Gy组、15Gy。并在2h后处死大鼠,收集大鼠腮腺及颌下腺组织。使用免疫印迹法检测Rad50蛋白在大鼠腮腺、颌下腺的表达情况;用TUNNEL法检测放射后大鼠涎腺组织的细胞凋亡情况。 结果:免疫印迹法检测显示,放射后大鼠在腮腺组织及颌下腺组织中的Rad50蛋白的表达量随着放射剂量的不同而改变;放射组与正常组比较,腮腺组织Rad50蛋白在9Gy(P0.05),颌下腺组织在12Gy(P0.05)达到最高。随着放射剂量的增加,Funel检测法检出阳性率增加。当放射剂量为9Gy(P0.05)时,腮腺组织中细胞阳性率最高;放射剂量为12Gy(P0.05)时,颌下腺组织中的阳性率最高。放射剂量对细胞凋亡阳性率有显著的影响,阳性率的变化与剂量相关。 结论:Rad50蛋白的表达水平受放射剂量和组织类型的影响,可能参与了涎腺放射损伤修复的机制。涎腺组织中腮腺的放射敏感性高于颌下腺组织。
[Abstract]:Aim: to study the changes of Rad50 protein expression in salivary gland of rats irradiated with different doses of C060 纬-ray and its effect on salivary gland apoptosis. Methods: sixty inbred male Wistar rats were randomly divided into 6 groups: control group (no radiation, 0Gy group), 3Gy group, 3Gy group, 12 Gy group, 15 Gy group, 10 rats in each group. The cumulative doses were 15 Gy in 3Gy group, 9 Gy in 9 Gy group and 12 Gy in 12 Gy group, respectively. The parotid gland and submandibular gland of the rats were collected after 2 hours. The expression of Rad50 protein in parotid gland and submandibular gland was detected by Western blot, and apoptosis in salivary gland was detected by TUNNEL assay. Results: Western blot assay showed that the expression of Rad50 protein in parotid and submandibular gland tissues changed with the dose of radiation, and the expression of Rad50 protein in the radiation group was higher than that in the normal group. Rad50 protein in parotid tissue was highest in 9Gy (P0.05), and in submandibular gland in 12Gy (P0.05). With the increase of radiation dose, the positive rate of Funel test increased. When the radiation dose was 9Gy (P0.05), the positive rate of cells in parotid gland was the highest; when the radiation dose was 12Gy (P0.05), the positive rate of submandibular gland was the highest. Radiation dose had a significant effect on the positive rate of apoptosis, and the change of the positive rate was correlated with the dose. Conclusion the expression of the protein is affected by radiation dose and tissue type, which may play a role in the repair of salivary gland injury. The radiosensitivity of parotid gland in salivary gland was higher than that in submandibular gland.
