大鼠尸体细菌群落演替与PMI推断意义

【摘要】:目的 本研究目的在于探索昆虫隔离环境中,大鼠自然体腔尸体腐败过程中细菌演替变化及规律,发掘尸体腐败期间细菌菌群及数量变化并评估利用细菌演替状况推断死亡时间(postmortem interval, PMI)的潜在价值。 方法 动物实验使用成年雌性SD大鼠,脊椎横断处死后置于隔离环境中,定期定点取样进行细菌培养,半定量计数及分离鉴定,分析不同时间点菌落组成及演替的变化情况。 结果 从时间点来说,葡萄球菌属和奈瑟菌属对早期死亡时间(Early PMI)有较好的指示作用,而乳杆菌属对晚期死亡时间(Late PMI)推断意义较强;从取样部位看,阴道的演替独立性和特殊性较其他取样点显示了更高的实用价值。 结论 1)大鼠尸体细菌群落演替随时间进展发生的变化具有规律性。 2)葡萄球菌属、奈瑟菌属对早期死亡时间具有指示作用。 3)乳杆菌属对晚期死亡时间具有指示作用。
[Abstract]:Objective to explore the variation and regularity of bacterial succession during the decay of rat natural body cavity in insect isolation environment. To explore the changes of bacterial flora and quantity during corrupting and to evaluate the potential value of estimating the time of death (postmortem interval, PMI) by bacterial succession. Methods female adult SD rats were used in animal experiments. The rats were killed by transection of vertebrae and placed in isolation environment. The bacteria were cultured by fixed point sampling, semi-quantitative counting and isolation and identification. The changes of colony composition and succession at different time points were analyzed. Results Staphylococcus and Neisseria had a better indication of early death time (Early PMI), while Lactobacillus had a stronger significance in predicting late death time (Late PMI). From the point of view of sampling site, the independence and particularity of vaginal succession showed higher practical value than other sampling points. Conclusion 1) the succession of bacterial communities in cadavers is regular over time. 2) Staphylococcus and Neisseria have indicative effect on early death time. 3) Lactobacillus can indicate the late death time.
