股四头肌创伤性骨化性肌炎的动物模型建立

【摘要】:目的:通过反复击打兔股四头肌的方法建立创伤性骨化性肌炎模型。方法:成年新西兰大白兔9只,以自体左后肢为模型组,右后肢为对照组。用钢球(质量:0.25 kg)以自由落体(高度100 cm)方式击打左后肢(模型组)股四头肌中部,随后每3天击打1次,第4周后停止击打,伸直位固定左膝关节;右后肢(对照组)同样方法固定。分别于第4、6、8周处死动物(均n=3),观察肿胀度、局部体征、局部骨化情况。结果:模型组股四头肌肿胀明显,关节逐渐强直、僵硬,局部触及硬结;对照组关节僵直,局部未触及硬结。影像学检查示模型组第4、6、8周击打处开始出现骨化影,对照组未出现。HE染色显示8周时击打处股四头肌肌肉组织内有明显类软骨或骨组织形成。结论:反复击打股四头肌的方法能成功建立单纯肌肉损伤导致的创伤性骨化性肌炎动物模型。
[Abstract]:Objective: to establish traumatic myositis ossificans model by beating quadriceps femoris muscle repeatedly. Methods: nine adult New Zealand rabbits were treated with autologous left hindlimb as model group and right hind limb as control group. A steel ball (mass: 0. 25 kg) was used to strike the middle of the quadriceps femoris of the left hind limb (model group) with a free fall (100 cm), then every 3 days. After 4 weeks, the hit stopped and the left knee was straightened. The right hind limb (control group) was fixed with the same method. The animals were killed at the 6th week of the 4th week (all nun3) to observe the degree of swelling, local signs and local ossification. Results: in the model group, the quadriceps femoris were swollen, the joints became stiff and stiff, and the joints in the control group were stiff, but not in the control group. Imaging examination showed that in the model group, the ossification began to appear at the striking place at the 4th week of the 6th week, but not in the control group. HE staining showed that there was obvious cartilage or bone formation in the muscle tissue of the quadriceps femoris muscle at the 8th week. Conclusion: the method of beating quadriceps femoris repeatedly can successfully establish the animal model of traumatic ossification myositis caused by simple muscle injury.
