晕船易感大鼠与不晕大鼠脑干蛋白质双向电泳图谱的鉴定与分析

【摘要】:目的:利用蛋白组学技术,建立晕船易感及不晕大鼠脑干总蛋白的双向电泳图谱,分析并鉴定差异表达蛋白质。方法:据模拟晕船刺激后异嗜高岭土量的增减将大鼠分为晕船易感组和不晕组,应用固相pH梯度(immobilized pH gradient,IPG)双向电泳(two dimensional electrophoresis,2 DE)技术,分离2 组大鼠脑干总蛋白,分析并用肽指纹图谱(peptide massfingerprint,PMF)鉴定差异蛋白质。结果:鉴定5个晕船易感相关蛋白质,上调酪氨酸3 单加氧酶/色氨酸5 单加氧酶激活蛋白、铁效应元件结合蛋白1,下调硫氧还蛋白过氧化物酶Ⅰ、磷酸甘油酸变位
[Abstract]:Objective: to establish a two-dimensional electrophoretic map of brain stem total protein in seasickness susceptible and non-dizzy rats by proteomics technique. Methods: according to the increase and decrease of heterophilic kaolin content after simulated seasickness stimulation, rats were divided into seasickness susceptible group and non halo group. Brain stem total proteins were isolated from two groups of rats and identified by peptide fingerprint peptide fingerprinting PMF. Results: five seasickness related proteins were identified, and tyrosine 3 monooxygenase / tryptophan 5 monooxygenase activator protein were upregulated.
