海水淹溺型急性肺损伤兔血管内皮生长因子及其受体的表达
[Abstract]:Objective to investigate the expression of vascular endothelial growth factor (VEGF) and soluble receptor-1 (sVEGFR-1) in serum and tissues after seawater drowning acute lung injury (SWD-ALI). Methods Twenty-eight healthy New Zealand rabbits were randomly divided into two groups: control group and model group (SWD-ALI model), which were observed at 3h and 6h time points (7 rabbits in each group), respectively. The pulmonary microvessel permeability index, lung wet / dry weight ratio and lung histopathological score were measured, the expression of VEGF and sVEGFR-1 in serum, alveolar lavage fluid (BALF) and lung tissue was detected by ELISA method, and the expression of mRNA in lung tissue was detected by RTPCR method. The distribution of VEGF in lung tissue was detected by immunohistochemical method. Results after seawater drowning, the wet / dry weight ratio of lung tissue, the permeability index of pulmonary microvessel increased, and the lung tissue pathological injury was serious. At 6 h, the wet / dry weight ratio of lung tissue was lower than 3 h, but the pathological score of lung tissue was still higher than 3 h. The levels of VEGF and sVEGFR-1 in serum increased significantly, and were positively correlated with the injury time (r = 0.837) and the pathological score (r = 0.796n / r = 0.839), and a positive correlation was found between the time of injury (r = 0.837) and the pathological score (P < 0.01). The expression of P0.01). VEGF and sVEGFR-1 in lung tissue and BALF was significantly decreased, and the expression of mRNA was also decreased, and then increased at 6 h. Immunohistochemistry showed that the expression of VEGF in the control group was large and evenly distributed, but after drowning, the expression of VEGF was significantly decreased and unevenly distributed. Conclusion the expression of VEGF and sVEGFR-1 in SWD-ALI is abnormal, and the changes of VEGF and sVEGFR-1 are consistent. The level of VEGF and sVEGFR-1 is related to the degree of lung injury.
