模拟失重下空间诱变菌感染巨噬细胞炎症相关microRNAs的筛选及生物信息学分析
[Abstract]:Objective to explore the differential expression of microRNAs (miRNAs) in macrophages infected with space mutated Escherichia coli (T1-13) under simulated weightlessness and its significance. Methods the second generation sequencing technique was used to detect the differential expression of miRNAs in macrophages of simulated weightlessness group and control group. The target genes of differential miRNAs were predicted by bioinformatics, and the GO function enrichment analysis was carried out on the target genes. KEGG signal pathway analysis and STRING protein interaction analysis. Results 33 differences in miRNAs, were down-regulated by 23 and increased by 10 between the simulated weightlessness group and the control group. These complementary miRNAs binding target genes are involved in MAPK pathway, WNT pathway, axon orientation and TGF- 尾 signaling pathway. The key down-regulated target gene is MAPK9,MAPK14,MAP3K7,MAP3K14,PIK3CA,PIK3CB,PIK3CD,MYC,JUN,SMAD4;. The key up-regulated target gene is AKT3,MAPK8,MAPK9,MAPK10,MAP2K1,PIK3CD. Conclusion there is a significant difference in miRNAs expression in macrophages infected by space mutagenesis under simulated weightlessness. These miRNAs may be involved in inflammatory reaction under weightlessness through regulating MAPK,WNT and TGF- 尾 pathways.
