白藜芦醇抗小鼠急性T淋巴细胞白血病作用及Notch1信号通路机制的研究

目的:观察白藜芦醇对急性T淋巴细胞白血病小鼠的作用及Notch1、Hes-1信号分子的表达水平,并探讨其作用机制。方法:1.完全随机法将50只C57BL/6雌性健康小鼠分为5组:正常对照组;模型对照组;白藜芦醇低浓度处理组(25mg/kg/d);白藜芦醇中浓度处理组(50mg/kg/d);白藜芦醇高浓度处理组(100mg/kg/d)。尾静脉注射1×106个GFP+白血病细胞,制备T-ALL白血病小鼠模型。2.应用瑞氏-吉姆萨染色观察各组外周血白血病细胞浸润情况。3.流式细胞术检测各实验组外周血、脾细胞悬液中白血病细胞的百分比。4.HE染色法观察各实验组肝脏、脾脏、肾脏、骨髓组织中白血病细胞的浸润情况。5.应用实时荧光定量PCR(RT-PCR)检测各组脾脏组织中Notch1、Hes-1信号分子mRNA的表达量。6.应用免疫蛋白印迹法(Western Blot)检测各组脾脏组织中Notch1、Hes-1蛋白表达水平。结果:1.模型对照组、白藜芦醇低、中、高浓度处理组的外周血涂片均可观察到原始淋巴细胞,其细胞核呈圆形或略呈卵圆形,大而深染,染色质致...
