淫羊藿苷经Wnt/β-catenin信号通路调控骨髓间充质干细胞的成骨—成脂双向分化

目的探讨Wnt/β-catennin信号通路介导的淫羊藿苷(Icariin)调控BMSCs(SD大鼠)成骨-成脂双向分化。方法1.体外实验(1)用细胞增殖、毒性试剂盒(CCK-8)检测BMSCs在多种不同浓度Icariin的干预其增殖活性的强弱及是细胞产生毒性反应。(2)用RT-q PCR确定Icariin对成骨分化指标(ALP、COL1、RUNX2)的影响,使用碱性磷酸酶(ALP)染色试剂盒和茜素红染色试剂确定经Icariin干预后ALP的活性及强弱、是否钙结节的形成及形成量(3)用RT-q PCR确定Icariin对成脂分化指标(PPARγ、Adipsin、FABP4)的影响,使用油红O染色试剂确定脂滴的形成。(4)使用信号通路特异性抑制蛋白DKK-1联合Icariin进行干预,检测Icariin在BMSCs成骨-成脂分化的作用。(5)Western-blot及Immunofluorescence检测Icariin干预后Wnt信号通路关键蛋白P-GSK、β-catennin表达和定位情况。2.在体试验用12周龄C57小鼠做卵巢切除模拟绝经后骨质疏松(OVX),口服Icariin(2...
