氧化苦参碱通过TGFβ1/P38/PAI-1信号通路逆转结肠癌上皮间质化及耐药的机制研究

目的:本课题研究拟以结肠癌细胞和构建结肠癌耐药细胞模型为基础,借助分子生物学及细胞生物学等先进技术,通过体外实验探讨氧化苦参碱对结肠癌细胞和结肠癌耐药细胞中TGF-β1/P38/PAI-1信号通路的干预作用,同时探索结肠癌上皮间质化(Epithelial-mesenchymal transition,EMT)对结肠癌耐药转移的作用机制,并以此来阐明氧化苦参碱逆转结肠癌EMT改变的分子机制及氧化苦参碱的抗肿瘤效用,为逆转结肠癌多药耐药提供新的研究方向。材料与方法:1.检测氧化苦参碱对于结肠癌细胞EMT的作用及迁移能力的影响。1.1对HT-29细胞和RKO细胞随机分组,以正常结肠上皮细胞FHC细胞作为对照组。使用Western blots方法分别检测各组细胞的EMT的相关标志物,包括上皮细胞标志物钙粘附蛋白E(E-cadherin)和间充质细胞标志物α平滑肌肌动蛋白(Alpha-smooth muscle actin,α-SMA)、纤连蛋白(fibronectin,FN)、纤溶酶原激活物抑制剂-1(Plasminogen Act ivat inhibitor-1,PAI-1)在内的蛋白表达...
