经股动脉注射途径建立前列腺癌和乳腺癌骨转移模型

【摘要】:目的探索建立高效、可靠的肿瘤骨转移动物模型的方法。方法将C57BL/6J雄鼠随机分为4组,每组15只;BALB/c雌鼠15只,设一组。将小鼠前列腺癌细胞RM-1和乳腺癌细胞4T1-luc消化、洗涤、计数后用生理盐水重悬至1×105/m L,取100μL,经股动脉将4T1 luc细胞注射到BALB/c雌鼠,并通过活体成像检验此方法转移成瘤的特异性。将100μL相同浓度的RM-1细胞悬液分别经股动脉、髂外动脉注射C57雄鼠各15只,比较两种不同注射方式所需时间、小鼠存活率,注射21天后将下肢骨HE切片,显微镜下观察并比较两种方法建立骨转移模型的成瘤率。结果经股动脉途径注射肿瘤细胞悬液所需的止血时间为2.53±1.75 min,经髂外动脉注射方法所需的止血时间为4.70±1.63 min(P0.05);经股动脉注射方法的手术时间为14.67±2.16 min,经髂外动脉注射方法的手术时间为22.47±3.50 min(P0.05);术后21 d经股动脉和髂外动脉注射组小鼠的生存率分别为93.3%和66.7%(P0.05);采用两种方法的肿瘤转移率均为100%;分别经股动脉和髂外动脉注射生理盐水组和肿瘤细胞悬浮液组在上述3项指标无显著差异。活体成像结果显示,肿瘤特异性地转移到了股骨和胫骨中。结论经股动脉注射方法建立肿瘤骨转移模型的手术及止血时间短、小鼠的存活率及肿瘤转移的成功率、特异性高,可作为肿瘤骨转移的新型动物模型。
[Abstract]:Objective to establish an efficient and reliable animal model of tumor bone metastases. Methods C57BL / 6J male mice were randomly divided into 4 groups, 15 female mice in each group were divided into one group. The mouse prostate cancer cell line RM-1 and breast cancer cell line 4T1-luc were digested and washed. After counting, 100 渭 L of normal saline was resuspended to 1 脳 105 / mL, and 4T1 luc cells were injected into the female BALB/c rat via femoral artery, and the specificity of the tumor was examined by in vivo imaging. The 100 渭 L RM-1 cell suspension of the same concentration was transferred to the femoral artery, respectively. 15 male C57 rats were injected into the external iliac artery. The time required for two different injection methods and the survival rate of the mice were compared. The lower extremity bone was sliced with HE on 21 days after injection. Results the hemostatic time for injection of tumor cell suspension through femoral artery was 2.53 卤1.75 min, and the hemostatic time for external iliac artery injection was 4.70 卤1.63 min (P0.05). The operative time of transfemoral and external iliac artery injection was 14.67 卤2.16 min and 22.47 卤3.50 min respectively, and the survival rate was 93.3% and 66.7% P0.05in femoral artery and external iliac artery injection group 21 days after operation, respectively. There was no significant difference in the above three indexes between the saline injection group and the tumor cell suspension group, and the results of in vivo imaging showed that there was no significant difference between the two groups. Conclusion the operative and hemostatic time of tumor bone metastasis model established by femoral artery injection is short, and the survival rate of mice and the success rate of tumor metastasis are high. It can be used as a new animal model of tumor bone metastasis.
