白细胞介素6对卵巢癌SOCS1和SOCS3表达的影响

【摘要】:目的:探究细胞因子信号抑制因子3(Suppressor of cytokine signaling protein 3,SOCS3)与白细胞介素6(Interleukin 6,IL-6)在卵巢癌组织中的表达及其临床意义,并通过将不同终浓度的IL-6作用于卵巢癌SKOV3细胞株,检测其对SKOV3细胞增殖能力的影响及SKOV3细胞中SOCS1和SOCS3表达的情况。方法:(1)采用免疫组化方法检测105例卵巢癌组织、40例卵巢良性肿瘤组织和30例卵巢正常组织中SOCS3和IL-6的表达,并使用相关的统计学方法分析两者的表达与卵巢癌临床病理参数的关系。(2)不同终浓度的IL-6对卵巢癌细胞SKOV3进行处理,行Cell Counting Kit-8(CCK-8)、逆转录聚合酶链反应(reverse transcription polymerase chain reaction,RT-PCR)、蛋白免疫印迹法(Western Blot)分别检测其对SKOV3细胞增殖能力的影响及细胞中SOCS1和SOCS3 mRNA和蛋白的表达。结果:(1)卵巢癌组织中SOCS3的阳性表达率显著高于良性肿瘤组织及正常卵巢组织(SOCS3:X2=57.672,P0.01),卵巢癌组织中IL-6的阳性表达率显著高于良性肿瘤组织及正常卵巢组织(X2=44.950,P0.01);SOCS3的过度表达与组织学分级及腹水有关(P0.05),其中,高级别且合并腹水的患者卵巢癌组织中的SOCS3阳性率显著高于低级别未合并无腹水的患者组织中的阳性率(P0.05);IL-6的过度表达与组织学分级、FIGO分期及腹水有关(P0.05),其中,晚期、高级别且合并腹水的患者的卵巢癌组织中的IL-6阳性率显著高于早期、低级别未合并腹水的患者组织中的阳性率(P0.05);Spearman相关分析表明SOCS3与IL-6的表达之间呈显著正相关(r=0.574,P0.05)。(2)不同终浓度IL-6处理SKOV3细胞后,SKOV3细胞的增殖水平具有浓度依赖性(F=3531.369,P0.01)。不同终浓度的IL-6作用于SKOV3细胞后,SKOV3细胞中SOCS1和SOCS3 mRNA和蛋白的相对表达量呈现浓度依赖性(F=18.832、261.112、6.096、58.922,P0.01)。结论:(1)SOCS3在卵巢癌组织中呈现高表达,且SOCS3的过度表达与组织学分级、及腹水有关,而与卵巢癌的组织类型、淋巴结是否转移、FIGO分期及年龄因素无关,其中,高级别且合并腹水的卵巢癌患者较低级别未合并腹水的卵巢癌患者相比,其癌组织中SOCS3的表达具有更高的阳性率。说明SOCS3的表达可能与卵巢癌的发生和发展具有密切的关联。IL-6在卵巢癌组织中呈现高表达,且IL-6的过度表达与组织学分级、FIGO分期及腹水有关,而与卵巢癌的组织类型、淋巴结是否转移及年龄因素无关。同时,晚期、高级别且合并腹水的卵巢癌患者较早期、低级别未合并腹水的卵巢癌患者相比,其癌组织中IL-6的表达具有更高的阳性率。这说明IL-6的过表达可能参与卵巢癌的形成,并且可能与卵巢癌病情进展有关。(2)随着IL-6浓度的升高,可抑制SKOV3细胞中SOCS1 m RNA及蛋白的表达,促进SOCS3 mRNA及蛋白的表达,这表明IL-6可能通过控制卵巢癌细胞中SOCS1及SOCS3表达的不同,调控卵巢癌细胞SKOV3的增殖。
[Abstract]:Objective: to investigate the expression and clinical significance of cytokine signal suppressor 3(Suppressor of cytokine signaling protein 3 (SOCS3) and interleukin 6(Interleukin 6 (IL 6) in ovarian cancer tissues, and to explore the clinical significance of the expression of cytokine signal suppressor 3(Suppressor of cytokine signaling protein 3 (SOCS3) and interleukin 6(Interleukin 6 (IL 6) in ovarian cancer SKOV3 cells. Methods the expression of SOCS3 and IL-6 was detected by immunohistochemical method in 40 cases of ovarian benign tumor and 30 cases of normal ovarian tissues, and the expression of SOCS3 and IL-6 in 10 5 cases of ovarian carcinoma and 30 cases of normal ovarian tissues were detected by immunohistochemistry. The relationship between the expression of SKOV3 and clinicopathologic parameters of ovarian cancer was analyzed by correlation statistical method. (2) the SKOV3 of ovarian cancer cells was treated with different final concentrations of IL-6. Cell Counting Kit-8 CCK-8, reverse transcription polymerase chain reaction- reverse transcriptase polymerase chain reaction (RT-PCR) and Western blotting were used to detect the effect of Cell Counting Kit-8 CCK-8 and reverse transcriptase polymerase chain reaction (RT-PCR) on the proliferation of SKOV3 cells and the expression of SOCS1, SOCS3 mRNA and protein respectively. The positive expression rate of IL-6 in ovarian cancer tissues was significantly higher than that in benign tumor tissues and normal ovarian tissues. The positive rate of IL-6 expression in ovarian cancer tissues was significantly higher than that in benign tumor tissues and normal ovarian tissues, and the overexpression of SOCS3 was related to histological grading and ascites. The positive rate of SOCS3 in high grade patients with ascites was significantly higher than that in low grade patients without ascites. The positive rate of IL-6 in ovarian cancer tissues of high grade patients with ascites was significantly higher than that in early stage. Spearman correlation analysis showed that there was a significant positive correlation between the expression of SOCS3 and IL-6 in patients without ascites. There was a significant positive correlation between the expression of SOCS3 and IL-6. (2) the proliferation level of SKOV3 cells treated with different final concentrations of IL-6 was different in a dose-dependent manner. The relative expression of SOCS1 and SOCS3 mRNA and protein in SKOV3 cells after treated with IL-6 at the final concentration was in a concentration dependent manner. Conclusion the expression of SOCS1, SOCS3 mRNA and protein in ovarian cancer tissues is high. The expression of SOCS1 and SOCS3 mRNA and protein in SKOV3 cells is higher than that in SKOV3 cells, and the expression of SOCS1 and SOCS3 mRNA and protein in SKOV3 cells is higher than that in SKOV3 cells in a dose-dependent manner. The overexpression of SOCS3 was related to histological grade and ascites, but not to the histological type of ovarian cancer, Figo stage of lymph node metastasis and age factors. Patients with high grade ovarian cancer with ascites compared with those with lower grade ovarian cancer without ascites. The positive rate of SOCS3 expression in human ovarian cancer tissues was higher. It suggested that the expression of SOCS3 might be closely related to the occurrence and development of ovarian cancer. IL-6 was highly expressed in ovarian cancer tissues. The overexpression of IL-6 was related to histological grade, Figo stage and ascites, but not to the histological type, lymph node metastasis and age factors of ovarian cancer. The positive rate of IL-6 expression in ovarian cancer tissues without ascites was higher than that in patients without ascites. This suggests that overexpression of IL-6 may be involved in the formation of ovarian cancer. And it may be related to the progression of ovarian cancer. (2) with the increase of IL-6 concentration, it can inhibit the expression of SOCS1 m RNA and protein, and promote the expression of SOCS3 mRNA and protein in SKOV3 cells. This suggests that IL-6 may control the different expression of SOCS1 and SOCS3 in ovarian cancer cells. To regulate the proliferation of ovarian cancer cell line SKOV3.
