诱导性胞嘧啶脱氨基酶表达与黑素瘤侵袭转移、预后及BRAF突变的相关性分析

【摘要】:目的:检测活性诱导性胞嘧啶脱氨基酶(AID)在黑素瘤中的表达,探讨其与黑素瘤侵袭转移、预后及BRAF突变的关系和临床意义。方法:AID蛋白在80例黑素瘤与23例色素痣石蜡包埋组织切片中的表达采用免疫组化SP两步法检测。使用spss数据分析软件分析蛋白表达情况结合临床特性、预后等的关系。结果:黑素瘤AID蛋白的阳性表达率53.75%(43/80),色素痣的表达率13.04%(3/23),差异有统计学意义(P0.05)。AID蛋白的表达与Clark分级与浸润深度呈正相关。伴有淋巴结转移的黑素瘤AID蛋白的表达率高。并且,AID蛋白阳性表达的黑素瘤预后差。(P0.05),不同年龄与性别的黑素瘤患者之间AID蛋白的阳性率差异不大。不同民族患者的蛋白表达率也没有较大差别(P0.05)。19例发生BRAF突变黑素瘤组织中AID蛋白17例阳性表达,其中15例BRAFV600E突变的黑素瘤AID蛋白均阳性表达。结论:AID蛋白可能可能诱导了黑素瘤BRAF突变,其参与黑素瘤的侵袭、转移并与预后密切相关,提示AID蛋白是一个潜在的预后判断指标。BRAF突变黑素瘤中AID蛋白高表达率为黑素瘤的靶向治疗提供了理论基础。
[Abstract]:Objective: to investigate the expression of active inducible cytosine deaminase (idd) in melanoma and its relationship with invasion and metastasis of melanoma, prognosis and BRAF mutation.Methods the expression of AID protein in paraffin-embedded tissue sections of 80 cases of melanoma and 23 cases of nevus pigmentosa was detected by immunohistochemical SP two-step method.Spss data analysis software was used to analyze the relationship between protein expression and clinical characteristics and prognosis.Results: the positive expression rate of AID protein in melanoma was 53.75% / 80%, and the expression rate of nevus pigmented was 13.04% / 23%. There was a positive correlation between the expression of AID protein and the grade of Clark and the depth of invasion.The expression rate of AID protein in melanoma with lymph node metastasis is high.The prognosis of melanoma with positive expression of AID protein was poor. The positive rate of AID protein in melanoma patients with different age and sex was not different.There was no significant difference in the expression rate of AID protein between patients from different nationalities and 19 cases of melanoma with BRAF mutation. There were 17 cases of positive expression of AID protein in melanoma tissues with BRAFV600E mutation, among which 15 cases were positive expression of AID protein in melanoma with BRAFV600E mutation.Conclusion the BRAF mutation of melanoma may be induced by the protein of: AID, which is involved in the invasion, metastasis and prognosis of melanoma.The results suggest that AID protein is a potential prognostic marker. The high expression rate of AID protein in BRAF mutant melanoma provides a theoretical basis for targeted treatment of melanoma.
